Immature persimmons are unripe fruits that are cut off during the persimmon cultivation process and immediately discarded, amounting to an annual fruit loss of approximately 100 to 400 kg per 1000 m2. The purpose of this study was to make effective use of unused resources, namely, immature persimmons, and attempt to use them as food additives. In this study, we studied the Tone Wase (fully astringent persimmon) and Fuyu (fully sweet persimmon) cultivars. As a result, we performed a component analysis of the immature persimmons, isolating 12 compounds, of which two were newly identified. Differences in the components and their contents were found between cultivars and between the peel and flesh. To effectively use immature persimmons as food for the elderly, we searched for active substances that inhibit AGE formation and found that extracts of immature persimmons and isolated compounds showed high activity. In particular, high activity was observed for catechin and its polymeric form, procyanidin. Regarding the inhibition of aroma deterioration, 5 mg/L of gallic acid in octadecane was found to be the optimal condition for the inhibition of citral deterioration. As for antimicrobial activity, we found that extracts at a concentration of 500 mg/L had no antimicrobial effect. Based on these findings, we made a microencapsulation process, and plan to advance to the clinical trial study in future. These findings confirmed the effectiveness of immature persimmons, which are an unused resource, and reveal their potential as a food for the elderly and as a food additive in other food products, which we hope will lead to new industrial innovations.

We evaluated the utility of gene-transcriptional responses in the liver of mosquitofish (Gambusia affinis), a species introduced to many countries and therefore widely available, for detecting endocrine-disrupting activity in water. Exposure to beta-naphthoflavone, an aryl hydrocarbon receptor (AhR) agonist, significantly increased the transcript of the cytochrome P4501A gene (cyp1a), peaking at 24 h, in both sexes at concentrations of 10 mu g/L or more. 17 beta-Estradiol (E(2)) at 500 ng/L increased the number of males showing gene transcription of precursors of yolk protein, vitellogenin (Vtga, Vtgb, and Vtgc), at 24, 48, and 72 h. Exposure for 48 h to bisphenol A (BPA), an estrogen mimic, also increased vtg-positive males at 1 mg/L or more. Leachate from a Japanese stable-type landfill significantly increased vtg-positive males after 48 h exposure, and the in vitro activity of the leachate against the estrogen receptor (ER) was estimated as an E(2) equivalent of 240 ng/L by yeast transfected with the ER. Chemical analysis showed that major contributors to the ER activation were BPA and 4-tert-octylphenol. This leachate and drainage water from a control-type landfill had AhR activities, estimated by yeast with the AhR, but had no significant effect on cyp1a transcription. These results showed that mosquitofish are suitable for detecting in vivo AhR and ER effects, but are insensitive to E(2). Environ. Toxicol. Chem. 2011;30:2778-2785. (C) 2011 SETAC

Seawater sample from the coral reefs of the Kerama Islands of Okinawa were assessed for the presence of carotenoid-producing bacteria. Results of 16S rDNA analysis of the bacteria obtained from the isolated bacteria showed unique patterns that were different from those of the bacteria obtained from the ordinary marine area. Phylogenetic analysis revealed a slight correlation with the statistical analysis of the PDA chart patterns. The results suggest that useful materials for human health such as carotenoids can be extracted from many carotenoid-producing bacteria such as those found the coral reefs the Kerama Islands.

The culture characteristics, carotenoid production, and associated biosynthetic pathway of strain T-1 were examined. As a result of examining the culture temperature and light irradiation, an increase of neurosporaxanthin and neurosporaxanthin β-D-glucopyranoside was observed at a low temperature and 0 lx. It was suggested that highly polar carotenoids, such as neurosporaxanthin, and carotenoid glycosides were involved in the stabilization of membrane during nutrition storage other than the defense function of fungus bodies. Strain T-1 produced lycopene, β-carotene, γ-carotene, torulene, neurosporaxanthin, and neurosporaxanthin β-D-glucopyranoside, as assessed by HPLC, LC-MS, and NMR analysis. Carotenoid biosynthesis begins with neurosporene, passing to lycopene and γ-carotene through cyclization, and produces β-carotene. In addition, it is saturated, γ-carotene is converted to torulene, and neurosporaxanthin is produced. Thus, the carotenoid biosynthetic pathway in strain T-1 was estimated.

Two resveratrol trimers, gnemonol B isolated from Gnetum gnemon and gnetin E obtained from the Gnetum species, were found to exhibit strong antibacterial activities against vancomycin-resistant Enterococci (VRE) and methiclllin-resistant Staphylococcus aureus (MRSA). The MIC values of gnemonol B against five strains of VRE and nine strains of MRSA were 12.5 and 6.25 μ g/ml, respectively. The MIC values of gnetin E against five strains of VRE and nine strains of MRSA ranged from 12.5 to 25 μ g/ml. These compounds also showed synergistic effects when used in combination with commercially available antibiotics according to the evaluation method using FIC indices. These findings suggested that the application of the test compounds alone or in combination with antibiotics might be useful in controlling and treating VRE and MRSA infections.

Three different extracts were separated by diethyl ether extraction, simultaneous steam distillation extraction and roasted aroma extraction (dry distillation) from Bursera graveolens. It was possible to identify 100 compounds in the three different extracts by GC-MS. The odour-active compounds present in the extracts were evaluated by gas chromatography-olfactometry (GC-0). It was estimated that mono- or sesquiterpenoids contributed to woody, herbal and minty aromas of the woody material of B. graveolens. On the other hand, the roast aroma produced by burning chips included several aroma compounds, such as cyclotene and vanillin. Copyright (c) 2005 John Wiley & Sons, Ltd.

Six phenolic compounds and one steroid were isolated from the leaves of Perilla frutescens Britton var. acuta (red perilla) with Japanese dietary pickled plum (Ume). These compounds were identified based on UV, MS, 1H-NMR and 13C-NMR spectroscopic data as β-sitosterol [1], shisonin [2], kaempferol 3-O-rutinoside [3], rosmarinic acid [4], rosmarinic acid methyl ester [5], 4-glucopyranosylcinnamic acid [6], and caffeic acid methyl ester [7]. In the bioassay, remarkable superoxide scavenging activity was found in the 1-butanol extract, and 20% methanol-water eluate, 50% methanol-water eluate and methanol eluate of an Amberlite XAD-2. In addition, remarkable superoxide scavenging activity was found in rosmarinic acid and rosmarinic acid methyl ester which are those isolation compounds. Moreover, we synthesized caffeic acid derivatives which were metabolic product of citrusin C and rosmarinic acid, and pharmacological effects such as superoxide scavenging activity used metabolite (vanillic acid, 4-hydroxy cinnamic acid, ferulic acid and isoferulic acid) of the chlorogenic acid as a starting material, and synthesized glycoside. And we examined their superoxide scavenging activity and tyrosinase inhibitory activity (Stepl as L-Tyrosine substrate Step 2 as L-DOPA substrate), and their synthesized compounds investigated for cosmetic use. As a result, for tyrosinase inhibitory activity, isoferulic acid (3,4-CA) and their methyl ester (3,4-CAM), good inhibition of 67.5 and 93.9% of Step 1 and 65.1 and 53.8% of Step 2 were noted. These compounds inhibited tyrosinase activity more than arbutin (63.0% of Step 1 and 7.7% of Step 2). For superoxide scavenging activity, ferulic acid (4,3-CA) and their methyl ester (4,3-CAM), and 4-hydroxy cinnamic acid (4-CA) were good activity of 43.6, 57.9 and 55.2%, respectively. Melanin inhibition and cytotoxicity were examined by using cultured B16 melanoma cells. More than 40% suppression was observed in 3-β-glucopyranosyl isoferulic acid methyl ester (3,4-CAGlc) and 4-hydroxy cinnamic acid methyl ester (4-CAM). © 2006, Japan Oil Chemists' Society. The characteristic spicy, sweet and balsamic aroma components of the woods of Bursera graveolens (palo santo) were measured using GC-MS. The characteristic four compounds, 1-4, were isolated from the diethyl ether extract. The structure of compounds 1 and 2 was elucidated by means of NMR studies, and confirmed to be 1-acetyl-4-isopropenyl-1-cyclopentene (1) and 2-methyl-5-isopropenyl-1-cyclopenten-1-carboxaldehyde (2) by synthesis from limonene oxide derivatives. The structure of compounds 3 and 4 was determined to be 6,10-epoxy-7(14)-isodaucane (3) and 10-hydroxy-6,10-epoxy-7(14)-isodaucane (4) by NMR experiments. Of these, compound 4 is a new sesquiterpenoid with a weak woody note, and the presence of compounds 1-4 in B. graveolens is reported here for the first time. A carotenoid biosynthesis gene cluster mediating the production of 2-hydroxyastaxanthin was isolated from the marine bacterium Brevundimonas sp. strain SD212 by using a common crtI sequence as the probe DNA. A sequence analysis revealed this cluster to contain 12 open reading frames (ORFs), including the 7 known genes, crtW, crtY, crtI, crtB, crtE, idi, and crtZ. The individual ORFs were functionally analyzed by complementation studies using Escherichia coli that accumulated various carotenoid precursors due to the presence of other bacterial crt genes. In addition to functionally identifying the known crt genes, we found that one (ORF11, named crtG) coded for a novel enzyme, carotenoid 2,2'-beta-hydroxylase, which showed intriguingly partial homology with animal sterol-C5-desaturase. When this crtG gene was introduced into E. coli accumulating zeaxanthin and canthaxanthin, the resulting transformants produced their 2-hydroxylated and 2,2'-dihydroxylated products which were structurally novel or rare xanthophylls, as determined by their nuclear magnetic resonance and high-performance liquid chromatography/photodiode array detector/atmospheric pressure chemical ionization mass spectrometry spectral data. The new carotenoid produced was suggested to have a strong inhibitory effect on lipid peroxidation.

Their structures were determined based on UV, MS, 1H-NMR and 13C-NMR spectroscopic data as kaempferol 3-O-rutinoside [1], kaempferol 3-O-glucopyranoside [2], quercetin 3-O-rutinoside [3], citrusin C [4], 2,3-dihydro-2-(4′-hydroxy-3′-methoxyphenyl)-3-β-D-glucopyranosylmethyl-7-hydroxy-5-benzofuranpropanol [5], corchoionoside C [6], and trans-carveol 6-O-β-glucopyranoside [7]. In the bioassay with superoxide scavenging activity, 1-BuOH extract, acidic components, neutral components, compound 3 and aglycone of 3 (3a) showed strong activity. And in the tyrosinase inhibitory activity, aglycone of 1 (1a) and aglycone of 3 (3a) showed good inhibition. As a result, we considered that Hibiscus sabdariffa may be related to the longevity, and their 1-BuOH extract was found promising as a cosmetic agent. The presence of compounds 4-7 in the leaves of Hibiscus sabdariffa is reported here for the first time.

To find a more efficient phytoremediation, the accumulation mechanism of elucidating heavy metals of a hyperaccumulator is important. We measured the behavior and glutathione quantity of the heavy metal accumulation level in calli using Brassica juncea, which is a hyperaccumulator of Pb and Cd. Brassica juncea from a seed was used to grow callus, which was for three months. Then, for two weeks, we conducted long-term exposure to a culture in heavy-metal component culture media (Cr, Cu, Zn, Cd, Pb, Co, Ni). Furthermore, we used a liquid culture medium for Pb, we conducted a short-term exposure for five days. As for Cd and Pb, the absorbed dose was high, and the Cd, in particular, showed the absorbed dose to be high at a low concentration (10-100 μM), compared with other heavy-metal classes. There were many things that necrotized, in other heavy metals, the fastness was not found to influence the heavy metal. In the short-term exposure of Pb, the absorption level increased during the early phase. However, 96 hours later, according to the cell cycle, that absorption appeared to be promoted. In addition, in GSH, while a large difference was not recognized, a high concentration level was much contained it. © 2005 The Japan Society for Analytical Chemistry.

In the course of studies on the constituents of the woody material of Bursera graveolens, the aroma components that provide its characteristic spicy, sweet and balsamic odor were investigated using GC/MS. Five compounds, 1-5, were isolated from the volatile oil obtained by diethyl ether extraction of B. graveolens wood chips, and their structures were elucidated by means of NMR studies. Their structures were determined as (-)-dihydro-α-agarofuran (1), (-)-5,11-epoxy-4α,5β,10α-eudesm-1-ene (2), (-)-4α-hydroxydihydroagarofuran (3), (-)-3β,4β -oxidoagarofuran (4) and (-)-10-epi-γ-eudesmol (5). Of these, (-)-5,11-epoxy-4α,5β,10α-eudesm-1-ene (2) is a new sesquiterpenoid that has not previously been reported in literature, and the presence of (-)-dihydro-α-agarofuran (1), (-)-4α -hydroxydihydroagarofuran (3), (-)-3β,4β-oxidoagarofuran (4) and (-)-10-epi-γ-eudesmol (5) in B. graveolens is reported for the first time. Atherosclerosis is a vascular disorder involving inflammation, a narrowed vascular lumen in the entire tunica intima, and reduced elasticity of the arterial wall. It has been found that Hsp60 from Chlamydia pneumoniae, an obligate bacterial pathogen associated with atheroma lesions, mimics human Hsp60, thereby causing attacks by immune cells on stressed endothelial cells expressing endogenous Hsp60 on their surface. Furthermore, Hsp60 from C. pneumoniae has been shown to promote the growth of vascular smooth muscle cells (VSMCs). To explore probes that can be used for studying signal transduction elicited by the chlamydial Hsp60, we have tested several natural products for their inhibitory actions on the Hsp60-induced proliferation of rat arterial smooth muscle cells. Sesamol, vanillyl alcohol, and trans-ferulic acid exhibited moderate inhibitory actions on the Hsp60-induced cell proliferation; zerumbone, humulene, and caryophylene effectively inhibited it at low concentrations with IC(50) values of 529, 122, and 110 nM, respectively. The results indicated that the 11-membered alicyclic ring is favorable for interactions with receptors involved in the Hsp60-induced VSMC proliferation.

PAHs are the general term of the compounds, having two or more benzene rings. These are discharged from diesel motor gas, tanker accidents, oil emissions by cars, and so on. They float in the atmosphere, and it is considered that they are absorbed in soil as a result of rain. Generally, compounds that have two or three benzene rings show only toxicity, whereas these having four or more benzene rings show toxicity, carcinogenicity and mutagenicity. Especially, benzo(a)pyrene was shown to be an agency of an endocrine disrupter. We investigated the action of 16 PAHs specified by the U.S. EPA in soil around Nara city. Soil was collected from different locations involving traffic and vegetation. Soil from three locations around our uni-versity were collected every month for investigating seasonal movement. PAHs were extracted from soil by soxhlet extraction with dichloromethane. They were then analyzed quantitatively by HPLC/UV. We classed PAHs by the number of rings, and examined the concentration and seasonal movements. All content of 16 PAHs in soils increased in proportion to traffic volume. At the same locations of traffic volume, the gravitation at a location with a plant with all 16 PAHs in soils was, furthermore, found to have a low concentration. There were different seasonal movements of the 2,3-ring and 4, 5, 6-ring PAHs. 4, 5, 6-ring PAHs have strong correlations each others(r > 0.79), but there was no correlation between 2, 3-ring and 4, 5, 6-ring PAHs. As a result, the traffic volumes are exposition sources of 4, 5, 6-ring PAHs in soils.

The surface of the sea in tropical and subtropical region is a severe environment for the growth of organisms, because active oxygen and free radicals are generated by intense irradiation with strong sunlight. Recently, it has been reported that the marine microorganisms Agrobacterium aurantiacum produced astaxanthin, which has one of the strongest ^1O_2 quenching activities. Then, focusing on fungal type marine microorganisms, we screened ocean fungus for production of excellent carotenoid with antioxidative activity. Here we report on the structural determination and cultural characteristic of a new carotenoid from marine microorganisms. Strain T-1 producing orange pigment was isolated from the surface of sea water at Tanegashima, Kagoshima Prefecture in Japan in August 1999 and identified as a Fusarium sp. The microorganism was cultured in a 10-liter scale at 25℃ for 7 days on a reciprocal shaker, using a modified marine broth medium. The cultured cells were concentrated by centrifugation, then extracted with acetone. After separation and purification by column chromatography on silica gel and HPLC, successively, structures of the carotenoids were analyzed using UV-VIS, MS, ^1H- and ^<13>C-NMR spectrometry. Neurosporaxanthin β-D-glucopyranoside was identified as a new carotenoid from the strain. A yield of carotenoid in 1g freeze-dried fungus body was found to be 0.25mg. Main carotenoids of the strain were neurosporaxanthin (45% out of the all carotenoids), γ-carotene (32%) and the new carotenoid neurosporaxanthin β-D-glucopyranoside (11%). β-Carotene (5%) and torulene (3%) were minor constituents.

When leaf segments of a tomato cultivar 'Ponderosa' were inoculated with Agrobacterium rhizogenes MAFF07-20001 carrying the binary vectors pRi and pBI121/sGFP, adventitious roots were developed from calli formed at the edges of the segments. Primordial roots were obtained with green fluorescence under blue light and elongated vigorously on hormone-free medium without loss of the green fluorescence. They were easily distinguishable from the non-fluorescing roots on the same segments. Successful integration of the sGFP and rol C genes into the chromosome of tomato roots was confirmed by polymerase chain reaction and Southern hybridization. The present method enables us to evaluate the hairy root formation without subculture, isolation and DNA analysis. All commercial cultivars available in Japan (24 cultivars) and 14 breeding lines of tomato were tested by this method. All but two breeding lines produced the hairy roots. Thus, the present method is useful for hairy root production in tomato. © 2003, Japanese Society for Plant Cell and Molecular Biology. All rights reserved.

Anacardic acids, (6-[(8Z)-8-pentadecenyl] salicylic acid (1) and 6-[(8Z,11Z)-8, 11,14-pentadecatrienyl] salicylic acid (2)) were isolated from the shell oil of Indonesian cashew nut and used to synthesize glycosides via glycosidation of the carboxyl group on the benzene ring. We examined these compounds for their inhibition of the enzyme tyrosinase, their superoxide scavenging activity and their inhibition of enzyme hyaluronidase. The glucosides derived from (1) and (2) were found to inhibit tyrosinase (using L-DOPA as a substrate) activity by more than 80%. The inhibitory rates exceeded that of arbutin, a compound that is used in commercial cosmetics. As the comparison, cardanols, (3-[(8Z)-8-pentadecenyl] phenol (3) and 3-[(8Z, 11Z)-8, 11, 14-pentadecatrienyl] phenol (4)) were isolated from the cashew nut shell oil and used to synthesize glycosides that we also examined for their inhibitory activity. © 2003, Japan Oil Chemists' Society. Six compounds were isolated from the leaves of Celosia argentea L. Tyrosinase inhibitory and superoxide scavenging activity of the compounds was examined so as to obtain a skin depigmentation agent for cosmetic application. Eugenyl Oβ-D-glucopyranoside (citrusin C, 1) showed strong tyrosinase inhibitory activity. Two-step inhibitory effects of successive tyrosine and DOPA oxidation for tyrosinase were stronger compared to arbutin used commercially at present (47.20% inhibition of tyrosine oxidation and 87.93%, DOPA oxidation, compared to 63.00% for tyrosine and 7.70% for DOPA oxidation in the case of arbutin). Moreover, we succeed in providing compound 1 abundantly in the synthetic study which used acetobromo-α-D-glucose and Sn(OTf)2-J. Oleo Sci., 51, 203-206 (2002).© 2002, Japan Oil Chemists’ Society. [Synopsis] Fast atom bombardment (FAB) mass spectrometry enables analyses in the positive and negative ion modes, providing different information. Therefore, the method is a powerful tool for analyzing the structure of polyisoprenepolyols. Although some of the structural information is available from routine FAB mass spectra, the linked scan spectra provides information in more detail. In this paper, we briefly review our recent works and describe the structural elucidation of polyisoprenepolyols isolated from an edible mushroom (Hypsizygus marmoreus) by FAB mass spectrometry. Pollution by chlorofluorohydrocarbons from freezers and air conditioners has recently contributed to the destruction of the ozone layer and it has been suggested that excessive exposure to ultraviolet rays causes serious problems such as carcinogenesis, inflammation, and photo-aging. The investigation of natural products have been increased expecting the effectiveness and safety. We have previously reported that 6-[(8Z)-8-pentadecenyl] salicylic acid (1) (Anacardic acid monoene) and 6-[(8Z, 11Z)-8, 11, 14- pentadeca trienyl] salicylic acid (2) (Anacardic acid triene) were isolated from cashew nut shell oil and their derivatives showed strong tyrosinase inhibitory activity. In this report, we synthesized eight derivatives from (1), (2), 3-[(8Z)-8-pentadecenyl] phenol (3) (Cardanol monoene) and 3-[(8Z, 11Z)-8, 11, 14-pentadecatrienyl] phenol (4) (Cardanol triene) by condensation and hydrolysis reactions using 2,3,4,6-tetra-o-acetyl-α-D-glucopyranosyl bromide and 2,3,4,6-tetra-o-acetyl-α-D-e galactopyranosyl bromide. These derivatives were examined for their use as whitening agents which inhibit tyrosinase activity, superoxide scavenging activity and hyaluronidase activity. Compounds (1a) and (2a) were found to be inhibited by ca. 81-88% tyrosinase activity (L-Dopa). This inhibitory action considerably exceeded that of arbutin used in commercial cosmetics.

[Synopsis] Four acetylenes and three terpenoids were isolated from Solidago altissima L. and characterized by MS, 'H-NMR and ^<13>C-NMR spectroscopy as methyl (Z)-decaene-4,6,8-triynoate (Zdehydromatricaria ester, 1), methyl 10-[(Z)-2-methyl-2-butenoyloxy]-(2Z,84-2,8-decadiene-4,6-diyoate (2), (4Z-2,4-decadiene-6,8-diyn-4-olide (Z-dehydromatricaria lactone, 3), (4E)-2,4-decadiene-6,8-diyn-4-olide (E-dehydromatricaria lactone, 4), 13E,7 α-acetoxyl kolavenic acid (solidagonic acid, 5), kolavenol (6), and D:C-friedours-7-en-3 β-ol (ilexol, 7). The polyacetylenes could be easily detected as yellowish brown spots on TLC plate following irradiation with blue black light for two to three minutes. The Z and E isomers of dehydromatricaria lactones were easily differentiated based on differences in melting points and chemical shifts of ^1H- and ^<13>C-NMR spectroscopy. Bioassay of these compounds on the germination of lettuce seeds showed that compounds 1, 2, 3, 4, and 5 exhibited growth inhibitory activity, whereas compound 7 did not exhibit the activity. Musk melon is the favorite fruit with a high market value in Japan, and the fragrance is one of the major factors determining the fruit quality of melon. In this study, mutant melon hairy roots which had been induced by means of the T-DNA insertion mutagenesis were found to produce volatile compounds with the fruity fragrance of mature melon. The volatile compounds were extracted and identified by GLC-mass spectrometry. Some essential oils such as (Z)-3-hexenol, (E)-2-hexenal, 1-nonanol, and (Z)-6-nonenol were stably synthesized by these hairy roots despite the increased number of subcultures. The productivity of these compounds by the best hairy root line was shown to be considerably higher than naturally ripened melon fruits.

Assuming that their foods may be related to the longevity, we started to investigate the constituents in the leaves of Celosia argentea L., which is used for diet in the country. Celosia argentea L. grows in the torrid zone of Africa by origin, and is also found in a warm place of the westward of Japan. Some terpenoids and saponins in the roots, and flavonoids in the leaves and stems have been implicated as active constituents in Celosia argentea L., Here we describe the isolation and identification of functional molecules in the leaves of African Celosia argentea L., and their effect on the skin depigmentation. Six compounds were isolated from the leaves of Celosia argentea L. and their structures were determined based on UV, MS, ^1H-NMR and ^<13>C-NMR spectroscopic data as 1-(4-O-β-glucopyranosyl-3-methoxyphenyl) propane-2-ene (citrusin C, 1), 3-O-β-glucopyranosyl-1H-indole (indican, 2), (3Z)-hexenyl-1-O-(6-O-α-rhamnopyranosyl-β-glucopyranoside) (3), (3Z)-hexenyl-1-O-β-glucopyranoside (4), (7E)-6,9-dihydromegastigma-7-ene-3-one-9-O-β-glucopyranoside (5), and trans-ferulic acid (6). Among them, compound 1 (citrusin C) showed a good skin depigmentation effect. The presence of compounds 1-6 in the leaves of Celosia argentea L. is reported here for the first time.

New polyisoprenepolyols (hypsiziprenol AA and BA) were isolated from an edible mushroom (Hypsizigus marmoreus). These polyols occur as a mixture of homologous polyisoprene derivatives with 40-70 carbon atoms. Analyses by FAB/MS in the positive and negative ion modes are complementary with each other in that the former provides information on the number of hydroxy groups present while the latter specifies the isoprenoid sequence, and thus become a powerful tool for analyzing the structures of polyisoprenepolyols. No polyisoprenepolyols obtained here were found to have antitumor activity on NCI-H292 and EL-4 cell lines.

Three limonoid glycosides were isolated from Citrus unshiu peels, and their structures were determined based on MS and NMR spectroscopic data as nomilinic acid 17-O-β-D-glucopyranoside (1), methyl nomilinate 17-O-β-D-glucopyranoside (2), and obacunone 17-O-β-D-glucopyranoside (3). In particular, the location of the sugar moiety was clearly determined by the B/E constant linked scan FABMS method. No limonoid glycosides obtained here were found to have antitumor activity in NCI-H292 and EL-4 cell lines. Copyright (C) 1999 Elsevier Science Ltd.

The structures of glycolipids isolated from Hypsizigus marmoreus (Bunashimeji, a mushroom) and Pleurotus citrinopileatus (Nireouma, a mushroom) were determined to be (4E, 8E)-N-2-hydroxyhexadecanoyl-1-O-beta-glucopyranosyl-9-methyl-C-18-sphinga-4,8-dienine (1), phosphodihexose N-(2-hydroxyoctadecanoyl)-4-hydroxy-C-18-(2), phosphodi-hexose N-(2-hydroxyhexadecanoyl)-4-hydroxy-C-18-sphinganine (3), phosphodihexose N-(2-hydroxytetracosanoyl)-4-hydroxy-C-18-sphi (4), and phosphodihexose N-(2-hydroxydocosanoyl)-4-hydroxy-C18-sphinganine (5). In particular, location of the double bonds in the lone-chain base of 1 was clearly determined by the B/E constant linked scan method. The structure of cerebroside having a long-chain base of 9-methyl-C-18-sphinga-4,8-dienine could be determined in general by the presence of characteristic fragment ions of [C-19-sphingadienine + H-H2O]+ at m/z 276 and [C-19-sphingadienine + H](+) at m/z 294, and the fatty acid carbon number could be calculated from the characteristic fragment ion of [ceramide-180](+) ([MH - GlcOH-180](+)) in positive ion mode FAB mass spectrometry. In the structural determination of 2-5, the ions of m/z 421 and 720 in the negative ion made analyses are assigned to be characteristic of phosphodihexose and phytosphingosine containing phosphodihexose, respectively This is a useful methodology for the structural determination of other unstable natural products such as lipids.

During the course of our search for functional molecules in edible fungi, we have reported the characterization and structural determination of the sphingolipids in Hypsizigus marmoreus (Bunashimeji, a mushroom) by B/E constant linked scan fast atom bombardment (FAB) mass spectrometry, that is, the fatty acid composition, the location of double bonds in the long-chain base (C19-sphingadienine), or the presence of phosphodihexose. In addition to these compounds, we also found many polyisoprenepolyols in a hot water-ethanol extract of an edible mushroom. In the polyisoprenepolyols, gymnopilins were discovered independently by two groups almost at the same time in 1983 (Nozoe et al. and Aoyagi et al.) from Gymnopilus spectabilis (o-waraitake, an hallucinogenic mushroom), as a bitter principle of the mushroom. However, gymnoprenol which has a free terminal hydroxy group instead of half-ester of 3-hydroxy-3-methylglutaric acid, has no bitter taste. Recently, Nozoe and Shirahama reported that these polyisoprenepolyols have an antitumor activity and also excitatory activity in central neurons. In the polyisoprenepolyols isolated from Hypsizigus marmoreus (Bunashimeji), we found new polyisoprenepolyols, which seem to be the precursor of gymnopilins. This compound has a terminal olefinic carbon group instead of a free terminal hydroxy group in gymnoprenol, and we named as hypsiziprenol. In the past, the structures of these polyisoprenepolyols were determined by chemical degradation studies, which require large amounts of sample. Here we describe, how information on the isoprenoid sequence and the number of hydroxy groups present can conveniently be derived by mass spectrometric methods that require only minute amounts of sample.

[Synopsis] Essential oils obtained from leaves, trunks, stalks, roots, and natural resins of plants, are the source of characteristic odours and consisted of terpenes, aromatic alcohols, aldehydes, ketones, esters, and so on. These compounds are present in cells of plant tissues and are mostly stored as glycosides. As occasion demands, these are discharged as odour in the atmosphere by the function of enzymes. In this manuscript, we have briefly reviewed some studies on odours, focussing on the plant odours and their chemical structures.

Four polyacetylenes including the Z and E isomers of dehydromatricaria lactones were isolated from Solidago altissima L. and characterized. The polyacetylenes could be easily detected as yellowish brown spots on TLC plate following irradiation with blue black light during two to three minutes. The Z and E isomers of dehydromatricaria lactones were easily differentiated based on differences in melting points and chemical shifts of 1H-and 13C-NMR spectroscopy. This study presents the first isolation of (E) -dehydromatricaria lactone from Solidago altissima L. in nature.

odern mass spectrometry has original advantages in that it enables observation of molecular signals even of labile compounds, by means of "soft ionization" techniques such as fast atom bombardment (FAB), and allows characterization of individual molecules present in a mixture with the use of tandem mass spectrometry (MS/MS method) or B/E constant linked scan method. These methods are successfully applied to the determination of natural organic compounds, in particular. In this manuscript, recent works are reviewed focussing on the structural elucidation of lipids including new glycosyl phosphosphingolipids isolated from mushrooms by FAB mass spectrometry.

A model study has demonstrated that phosphorylation of a highly hindered hydroxyl group can be achieved via sequential treatment with (1) 2-trimethylsilylethyl dichlorophosphite, (2) 2-trimethylsilylethyl alcohol, and (3) hydrogen peroxide. The resulting triester survives a variety of reaction conditions but may be cleaved with fluoride ion to afford the phosphate monoester.

We report here the difference between phenyl propanoid glycosides obtained from hot water and methanol extracts of lemon peel. Eight phenyl propanoid glycosides were isolated, and their structures were established by UV, MS, H-1-NMR and C-13-NMR spectroscopy, as well as by chemical evidence. They were coniferin (1), syringin (2), dehydrodiconiferyl alcohol-4-beta-glucoside (3), citrusin A (4), citrusin B (5), citrusin C (6), methyl-3-(4-beta-glucopyranosyl-3-methoxyphenyl)propionate (7) and methyl-3-{4-(6-O-alpha-glucopyranosyl-beta-glucopyranosyl)-3-hydroxyphenyl}propionate (8). Compound 7, after intravenously injecting (0.5 mg/100 g of body weight) into stroke-prone spontaneously hypotensive rats (SHR-SP), was found to lower the blood pressure. Among these isolated compounds, 7 and 8 are new phenyl propanoid glycosides.

As a series of our studies on physiologically active substances in citrus fruit peels, we report here the differences between flavonoid glycosides in Citrus unshiu peelings (the ripe fruit peels) and flavonoid glycosides in young Citrus unshiu peelings (the unripe fruit peels). Eleven flavonoid glycosides were isolated from young Citrus unshiu peels and their structures established by UV, MS, 1H-NMR and 13C-NMR spectroscopy and also by chemical findings. They were limocitrin 3-β-D-glucoside (1). narirutin (4), rutin (5), narcissin (6), hesperidin (7), 3-hydroxy-5, 6, 7, 8, 3', 4'-hexamethoxyflavone 3-β-D-glucoside (8), 6, 8-di-C-glucosylapigenin (11), 3, 8-di-C-glucosylapigenin (12), 3, 7, 4'-trihydroxy-5, 6, 8, 3'-tetramethoxy flavone 3-β-D-glucoside (13), 3, 7, 4'-trihydroxy-5, 6, 8, 3'-tetramethoxyflavone 3-O- {[3-hydroxy-3-methylglutaryl (1→6)] -β-D-glucoside} (14) and limocitrin 3-O- {[3-hydroxy-3-methylglutaryl (1→6)] -β-D-glucoside} (15).
Five flavonoid glycosides (11) (15) were found not to be present in Citrus unshiu peelings (the ripe fruit peels), but could be isolated from young Citrus unshiu peelings (the unripe fruit peels).
Thus the kinds and quantity of flavonoid glycosides in ripe fruit peels and those in unripe fruit peels differ greatly. The relationship between their structures and hypotensive activity was determined by injecting the compounds intravenously in SHR-SP.
Of these compounds, (13), (14), and (15) were identified here for the first time as flavonoid glycosides.

During the course of our studies on physiologically active substances in citrus fruit peels, 27 compounds (phenyl propanoid glycosides, terpenoid glycosides, limonoid glycosides and adenosin) were isolated from lemon, unshiu, grapefruit, kinkan, sudachi, yuzu, hassaku, zabon, orange, iyokan and amanatsukan peels. On investigating phenyl propanoid glycosides which are expected to have seful physiological activity from a structural view point, we isolated 7 compounds including 4 new glycosides, providing interesting information with regard to physiological activity. As for terpenoids, 7 terpenoids and 7 terpenoid glycosides were isolated and their structures and bioassay were investigated. We also isolated 5 limonoid glycosides. Interestingly, we found in several citrus fruit peels, in the presence of adenosin, which exhibited the strongest hypotensive effect. Among these 27 compounds isolated, 13 are hitherto unreported compounds and the rest have been isolated from citrus fruit peels for the first time.

The structure and hypotensive effect of phenyl propanoid glycosides in hassaku and orange peels are described in a series of studies of physiologically active substances in citrus peels. Six phenyl propanoid glycosides were isolated, and their structures were established: coniferin[l], syringin[2], dehydrodiconiferyl alcohol-4-α-glucoside[3], 1-(4-glucosyl-3-methoxyphenyl)-2-{2-methoxy-4-[1-(E)-propene-3-ol]-phenoxy}-propane-1, 3-diol[4], i-(4-glucosyl-3-methoxyphenyl)-2-{2,6-dimethoxy-4-[1-(E)-propene-3-ol]-phenoxy}-propane-1,3-diol [5] and 1-(4-glucosyl-3-methoxyphenyl)-propane-2-ene [6] by MS, UV, 1H-NMR and 13C-NMR spectroscopy and chemical evidence. We propose the terms citrusin A, B and C for these new phenyl propanoid glycosides([4], [5] and [6]). Each component was intravenously injected in SHR-SP rats (1mg/100g body weight), and compounds [1] and [2] were found to raise rat blood pressure, but compounds [3], [4] and [6] lower rat blood pressure. © 1986, Japan Society for Bioscience, Biotechnology, and Agrochemistry. A search was made to isolate hypotensive substances from the hot water extract of citrus fruits peels. The forty-four flavonoid glycosides were isolated by repeating chromatography and by gel filtration after extracting with butanol and treating with lead subacetate. Their structures were established by MS, UV, ^1H-NMR and ^<13>C-NMR spectroscopy and the sugar analysis. Among these compounds, the nineteen flavonoid glycosides were new. Each component was intravenously injected in SHR-SP(0.3〜1mg/100g of body weight), and many compounds were found to lower the rat blood pressure. In especial, hypotensive effect of 6,8-di-C-glucosylapigenin showed the effect very strongly.

We compared the official method using AAS or with our new quick heavy metal measuring system based on voltammetry method. Then we considered whether analysis of cadmium on paddy-rice investigation was possible. As for the measurement of heavy metals by microcartridge type electrochemistry analysis machine, measurement was possible in lower than 10 ppb which was Japanese reference value easily, and manipulation was simple and easy. Also we examined the influence that we used a pumpkin as a clue of a heavy metal detoxication mechanism by this study, and Mn presence gave glutathione dosage in one of the defence materials in Pb accumulation dosage in Pb exposition and microelement and organism. As a result, Mn presence affected rhizoma interlobar Ca distribution. In addition, Mn free experiment area showed a rise of GSH/GSSG rate in leaf region conspicuously. In Pb concentration(400 ppm), it compared it with Mn presence experiment area in Mn free experiment area and decreased conspicuously. From these results, it was suggested that Mn functions for an action of a glutathione-related enzymatic group.
