    Headspace solid-phase microextraction combined with GC/MS (HS-SPME-GC/MS) is one of the strongest tools for comprehensive analysis of volatile compounds and has been used to analyze aromatic components of mango and investigate its varietal characteristics. In this study, profiling of aroma compounds in 17 mango cultivars, grown in the same green house to exclude the effect of environmental factors, was conducted and the patterns were subjected to principal component analysis (PCA) to identify the relationship between the aroma components and cultivars. Fifty-nine different volatile constituents were detected from the blends of these 17 mango cultivars. The cultivars were divided into four clusters using PCA based on the volatile components determined in the study. Aiko was found to mainly contain δ-3-carene and showed a composition more similar to its pollen parent, Irwin, than to its seed parent, Chiin Hwang No. 1.

    This study examines the effect of quantity sold (sales volume) on profitability of market participating smallholder farmers in northern Ghana. Market participation has been shown to be important for increasing incomes and improving production efficiency for farm households but still remains low in SSA. While agribusiness and development experts generally advocate for more intensive market participation, it is not clear if selling more results in more profits for smallholder farmers in remote markets that are prone to exorbitant transaction costs. The data used in this study is from the APS survey conducted in 2013 and 2014 in Northern Ghana which had a sample size of 527. The study is based on the theory of profit maximization, in which separability is inferred from observed market participation. OLS regression is used for empirical estimation after rejecting the hypothesis of endogeneity in the model. Mean gross margin/ kg across four groups of farmers ranked by quantity sold is also statistically examined. The results confirm the existence of economies of scale and also show that different crops have different effects on profitability. The results also show that although unambiguously positive, the relationship between quantity sold and profitability may not be linear.

    During the course of Citrus unshiu fruit cultivation, large amounts of plant material including pruned leaves, thinned-out flowers and unripe fruit are obtained; these materials are generally considered unusable and discarded as plant wastes. We have previously attempted to establish an effective use of such plant wastes as beneficial natural resources and found that a methanolic extract of pruned Citrus leaves (CUL-ext) exhibited inhibitory activity against porcine pancreatic lipase. In this study, we sought to identify further the effective uses of C. unshiu plant wastes by determining the lipase inhibitory activity of methanolic extracts of thinning out flowers (CUF-ext) and unripe fruit (CUUF-ext). We accordingly found that the inhibitory activity of CUF-ext was superior to that of CUUF-ext and comparable to that previously observed using CUL-ext. Fractionation of CUF-ext and CUUF-ext, followed by chromatographic analyses, revealed that the pancreatic lipase inhibitory activities of these extracts could be attributed, at least in part, to the flavonoids hesperidin, nobiletin, narirutin and rutin. On the basis of present findings, we propose that, in addition to pruned Citrus leaves, the thinned-out flowers and unripe fruit of C. unshiu are natural resources which are suitable for preparing constituents with lipase inhibitory activity.

    Plants produce ∼300 aromatic compounds enzymatically linked to prenyl side chains via C-O bonds. These O-prenylated aromatic compounds have been found in taxonomically distant plant taxa, with some of them being beneficial or detrimental to human health. Although their O-prenyl moieties often play crucial roles in the biological activities of these compounds, no plant gene encoding an aromatic O-prenyltransferase (O-PT) has been isolated to date. This study describes the isolation of an aromatic O-PT gene, CpPT1, belonging to the UbiA superfamily, from grapefruit (Citrus × paradisi, Rutaceae). This gene was shown responsible for the biosynthesis of O-prenylated coumarin derivatives that alter drug pharmacokinetics in the human body. Another coumarin O-PT gene encoding a protein of the same family was identified in Angelica keiskei, an apiaceous medicinal plant containing pharmaceutically active O-prenylated coumarins. Phylogenetic analysis of these O-PTs suggested that aromatic O-prenylation activity evolved independently from the same ancestral gene in these distant plant taxa. These findings shed light on understanding the evolution of plant secondary (specialized) metabolites via the UbiA superfamily.

    In this study, we aimed to identify the utility of pruned mango (Mangifera indica ‘Irwin’) leaves as a resource for ingredients with antioxidant activity. Firstly, we examined the antioxidant activity of extracts obtained from the pericarps, flesh, flowers, barks, seeds, young dark reddish brown leaves (YDL-ext), young yellow leaves (YYL-ext), and pruned old dark green leaves (OML-ext) obtained from ‘Irwin’ mango. Among them, methanolic extract of flower and OML-ext showed the most potent 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and superoxide dismutase (SOD)-like activity. The flesh extract showed weak DPPH radical scavenging activity, but did not show SOD-like activity. Secondly, we investigated the relationship between the maturation of leaves and their antioxidant activity by considering the contents of their two active polyphenolic components, 3-C-β-D-glucosyl-2,4,4’,6-tetrahydroxybenzophenone (1) and mangiferin (2), in addition to chlorophyll (3) and anthocyanins represented by cyanidin-3-O-glucoside (4). The DPPH radical scavenging activity of YDL-ext, YYL-ext and OML-ext were mainly attributable to 1, 2 and 3, whereas their SOD-like activity was partly attributable to 2. The DPPH radical scavenging and SOD-like activities of YDL-ext and YYL-ext were attributable to 1 and 2. These activities were also due to anthocyanins whose content is highest in YDL-ext. Considering the amounts of leaves obtained from pruning, old dark green leaves may be a reasonable natural resource for preparing cosmetics and/or supplemental ingredients with health-enhancing properties, antioxidant activity and inhibitory effect on AGEs formation and pancreatic lipase.

    As a part of our ongoing research to find novel functions in mango leaves, we have reported that the methanolic extract of pruned old dark green mango leaf (Mangifera indica ‘Irwin’) exhibited inhibitory effects on the formation of advanced glycation end products (AGEs) in nonenzymatic glycation of albumin. The purpose of this study was to find other mango cultivars with more potent activity in this regard. We examined the inhibitory effect of seventeen mango (Mangifera indica) cultivar leaf extracts on AGEs formation. We also investigated the relationship between the inhibitory activity of the extracts and the contents of their active components, 3-C-β-D-glucosyl-2,4,4’,6-tetrahydroxybenzophenone (1), mangiferin (2) and chlorophyll (3). On the basis of the evaluation of the inhibitory activity of mango cultivar leaf extracts, the HPLC determination of the contents of 1 and 2, and the spectrophotometric determination of 3, it was found that almost all extract showed a significant activity, and the content of 2 and 3 detected in each was similar. In contrast, AGEs formation inhibition tended to be higher as the content of 1 in the leaf extracts increased. This is the first report of phytochemical analysis of compounds 1, 2 and 3 in various cultivars of mango leaf. From the phytochemical point of view, these results suggest that the pruned leaves of any cultivar of Mangifera indica except ‘Chiin Hwang No. 1’ and ‘Kyo Savoy’ may be useful for the preparation of natural ingredients with inhibitory activity of AGEs formation.

    In the cultivation of Citrus fruit, pruning in late winter after harvest is important to obtain excellent flower buds in the next spring and a rich harvest of superior fruit in the following winter; a large number of pruned leaves are usually discarded. The purpose of this study was to determine the utility value of pruned leaves of Citrus unshiu. A methanol extract of pruned C. unshiu leaves showed a porcine pancreatic lipase inhibitory activity. Fractionation of C. unshiu leaf extract (CUL-ext) followed by bibliographic and chromatographic analyses revealed that a part of the pancreatic lipase inhibitory activity of CUL-ext was attributable to nobiletin (1), rutin (2), and hesperidin (3). Thus, pruned C. unshiu leaves may be a reasonable natural resource for the preparation of ingredients with lipase inhibitory activity.

    The purpose of this study is to isolate the beneficial microorganisms whose growth is promoted in the presence of charcoal materials. We successfully isolated strain IA, whose growth is promoted on an agar plate with charcoal materials, and identified it as a novel strain of the Bacillus sp. The growth of strain IA in the liquid medium was promoted by the addition of both activated charcoal (AC) and rice husk biochar (RHB). Moreover, the sporulation of strain IA in the RHB medium and the antifungal activity of the culture supernatant of the RHB medium were much higher than those with AC. HPLC and MS analyses revealed that strain IA produced an antifungal lipopeptide iturin A, and the yield of iturin A in the RHB medium was 8 times higher than that in the medium without RHB. This is the first paper to describe the positive effect of RHB on microbial metabolisms.

    Mechanical damage is one of the unavoidable environmental stresses to plant growth and development. Plants induce a variety of reactions which defend against natural enemies and/or heal the wounded sites. Jasmonic acid (JA) and salicylic acid (SA), defense-related plant hormones, are well known to be involved in induction of defense reactions and play important roles as signal molecules. However, defense related metabolites are so numerous and diverse that roles of individual compounds are still to be elucidated. In this report, we carried out a comprehensive analysis of metabolic changes during wound response in citrus plants which are one of the most commercially important fruit tree families. Changes in amino acid, sugar, and organic acid profiles in leaves were surveyed after wounding, JA and SA treatments using gas chromatography mass spectrometry (GC/MS) in seven citrus species, Citrus sinensis, Citrus limon, Citrus paradisi, Citrus unshiu, Citrus kinokuni, Citrus grandis, and Citrus hassaku. GC/MS data were applied to multivariate analyses including hierarchical cluster analysis (HCA), primary component analysis (PCA), and orthogonal partial least squares-discriminant analysis (OPLS-DA) to extract stress-related compounds. HCA showed the amino acid cluster including phenylalanine and tryptophan, suggesting that amino acids in this cluster are concertedly regulated during responses against treatments. OPLS-DA exhibited that tryptophan was accumulated after wounding and JA treatments in all species tested, while serine was down regulated. Our results suggest that tryptophan and serine are common biomarker candidates in citrus plants for wound stress. (C) 2016, The Society for Biotechnology, Japan. All rights reserved.

    gamma-Oryzanol is a main oleophilic component in rice bran oil and has been well recognized as a good dietary supplement for human health, as well as having uses in industrial materials. japonica-type rice cultivars generally showed significantly higher contents of total gamma-oryzanol in brown rice compared with indica-type cultivars, although within-group variation was significant. The objective of this study was to explore quantitative trait loci (QTLs) responsible for the difference in the gamma-oryzanol content between japonica-type and indica-type rice cultivars, using recombinant inbred lines (RILs), backcross inbred lines (BILs), and corresponding chromosome segment substitution lines (CSSLs) derived from crosses between japonica-type and indica-type. Results from RILs and BILs showed that eight QTLs were detected with R-2 from.09 to.16. Nine candidate regions for QTL were also suggested from corresponding CSSLs. These QTLs from RILs and BILs and the candidate regions from CSSLs were not overlapped, although one QTLs was mapped near the boundaries of the respective candidate region. At four QTLs and three candidate regions, alleles or segments from japonica-type caused higher contents than those from indica-type. On the other hand, at the other four QTLs and six candidate regions, alleles or segments from indica-type caused higher contents than those from japonica-type, which is a reverse result to the parental differences. This result strongly suggested that alleles with increasing effects on.-oryzanol content could be accumulated not only from japonica-type but also from indica-type, leading to a potential for increase in gamma-oryzanol content in future breeding programs.

    果実の追熟条件の違いが,ウメ '露茜' 果実のアントシアニンの蓄積や果実成分含量に及ぼす影響について調査した.エチレンを添加して20°Cで追熟した場合,果皮を含む果肉のアントシアニン含量は,追熟後4日頃より急増した.エチレンを添加して同じ期間追熟した場合,アントシアニン含量は20°Cおよび25°Cが15°Cおよび30°Cに比べて高かった.エチレンを添加すると追熟中にアントシアニン組成が変化した.赤く色づき始める前に収穫された果実では追熟後の赤色着色が不十分であり,適熟で収穫した果実ではそれより前に収穫した果実に比べて,追熟後のアントシアニン含量が低かった.追熟により果実全体が着色した果実では,大きさの違いはアントシアニン含量に影響しなかった.アントシアニン以外の果実成分については,リンゴ酸が追熟前後ともに,採取日が遅くなるほど高くなった.また,ポリフェノール含量は,追熟により高くなった.結論として, '露茜' 果実の収穫時期や追熟温度により追熟後のアントシアニン含量が大きく変わることが確認された.アントシアニン含量を効率よく高めるためには,果実全体が色づく適熟期よりもやや未熟な,果実表面の3~5割着色した果実を収穫して,エチレンを添加し,20°Cで4日程度追熟させるのが適当と考えられた.

    Citrus plants are world widely cultivated as horticultural tree crops, and nowadays their pharmacological activities have been well studied. Since research of defense responses in citrus plants have been mainly focused on the post-harvested fruits because of their commercial importance, defense mechanisms during their developmental stages have not been well understood. In the present study, two wound-induced compounds were isolated from leaves of Citrus hassalcu, and their structures were elucidated by high-resolution electron spray ionization mass spectra (HRESIMS) and nuclear magnetic resonance (NMR) analyses. One of these compounds was identified as a known flavanone, hesperetin. The other was characterized as a novel furofuran lignan, and was named `biscitrusnin-A'.     Citrus plants are well known as a rich source of VOCs, and several have important roles in defense responses. However, how VOCs are regulated in response to environmental stress is not yet well understood. In this study, we investigated dynamic changes of VOCs present in leaves of seven Citrus species (Citrus sinensis, C. limon, C. paradisi, C. unshiu, C. kinokuni, C grandis, and C hassaku) in response to mechanical wounding, jasmonic acid (JA), and salicylic acid (SA) as determined by gas chromatography/mass spectrometric analysis followed by multivariate analysis (principal component analysis, PCA, and orthogonal partial least squares-discriminant analysis, OPLS-DA). PCA and OPLS-DA suggested that changes in VOC profiles against stress stimuli were much diverse among Citrus species. OPLS-DA showed that C6 volatiles, such as hexanal and trans-2-hexenal, were induced in response to JA and SA stimuli in C sinensis and C. grandis, while the other VOCs were decreased under all tested stress conditions. alpha-Farnesene was induced in all species except C. hassaku after wounding or JA treatment. In addition, alpha-farnesene was also induced in response to SA stimuli in C. unshiu and C kinokuni. Therefore these volatiles can be candidates of the common stress biomarkers in Citrus.     The utilization efficiency of solar energy through photosynthesis by crops is only a few percent at most. Therefore, most solar energy in the field (on-site) remains unused. Plants are, however, able to use certain types of monochromatic light as an environmental signal, and they exhibit various physiological responses to even small amounts of the light. In particular, the significant induction of secondary metabolite production by blue light is well documented. Here, we propose a novel cropping method where unused solar energy is captured on-site by solar cells and converted to monochromatic light by using LEDs. The crops are then irradiated to stimulate enhanced production of secondary metabolites, thus improving crop function without the need for an external energy source. In this study, the effect of blue light on the production of functional secondary metabolites was evaluated in perilla (Perilla frutescens var. crispa f. purpurea), a traditional medicinal crop in Japan. Perilla leaves contain several functional substances such as perillaldehyde, rosmarinic acid, and anthocyanins, which have notable stomachic, antipyretic, detoxification, sedation, and diuretic efficacy. Plants were grown under natural daylight supplemented with blue light irradiation (460 nm, approx. 10 mu mol/m(2)/s, 24 h,) for 108 days before harvest. Control plants were grown without supplemental lighting. The content ratio of perillaldehyde in plants grown with blue light was 60% higher at harvest than in the controls. Moreover, in the vegetative growth stage, the perillaldehyde content increased significantly after only three days of blue light irradiation. These findings suggest that the present approach will benefit agricultural practices in developing countries through more efficient usage of on-site solar energy. (C) 2014 The Authors.     紀の川流域で古くから栽培されてきた地域遺伝資源としてシソの1系統を見出した.本研究では,まず,本系統の葉の形態および機能性香気成分の組成比における特徴を4種のシソ品種と比較した.その結果,本系統は,葉の両面が赤紫色で縮緬状の起伏がなく,ペリルアルデヒドとリモネンを多量に含むことから,薬用のアカジソ(P. frutescens var. crispa f. purpurea)の1種と推察された.一方で,本系統の特産品化を目指して,新規用途の開拓と周年生産に向けた栽培技術の検討を進めた.自然光下で栽培されているシソに青色光補光を行うと,花成が抑制されて若い葉が分化し続けた.この結果として,ペリルアルデヒド含有量を高く維持することができた.この方法は,短日期のシソの花芽形成を抑制し,機能性成分含有量を向上させる上で有用である.

    Citrus is one of the most important fruit trees in the world and spreads to temperate, sub-tropical and tropical areas. From ancient times Citrus species have been used as fresh fruits, cooking, medicines and ornamentals in Japan. Although the origin and time of introduction is unclear, many old species such as Tachibana (Citrus tachibana), Koji (C. leiocarpa), Shekwasha (C. depressa), Daidai (C aurantium), Yuzu (C. junos), Kabosu (C sphaerocarpa) have been traditionally cultivated in Japan. Kishu-mikan (C. kinokuni) was one of the earliest species commercially cultivated in Japan during Edo era. Satsuma mandarin (C. unshiu) originated in Kagoshima prefecture, southern part of Kyushu Island, as a chance seedling of some Citrus fruit or form imported from China and has been the main Citrus species to support the modern citrus industry of Japan. More than 200 cultivars have been developed by bud mutation and breeding program. In addition, some species of Citrus, such as Natsudaidai (C. natsudaidai), Hassaku (C. hassaku), Iyo-kan (C. iyo), Hyuganatsu (C. tamurana), Sanboukan (C. sulcata) and pummelo cultivars originated in Japan. Indigenous and old species have been cultivated and maintained in limited areas as local products with small consumption. On the other hand, many species have disappeared due to the low economical value. Indigenous and old cultivars adapted to the Japanese climate condition for a long time have the potential as materials for breeding program. Kinki University maintained indigenous and old Citrus species for phylogenetic studies to pursue the origin of species. Many old species are also maintained in China or Korea. Regional networks in these countries will be required for germplasm conservation of Citrus species.

    The wheat ( Triticum aestivum) and rye ( Secale cereale) beta-D-glucosidases hydrolyze hydroxamic acid-glucose conjugates, exist as different types of isozyme, and function as oligomers. In this study, three cDNAs encoding beta-D-glucosidases (TaGlu1a, TaGlu1b, and TaGlu1c) were isolated from young wheat shoots. Although the TaGlu1s share very high sequence homology, the mRNA level of Taglu1c was much lower than the other two genes in 48- and 96-h-old wheat shoots. The expression ratio of each gene was different between two wheat cultivars. Recombinant TaGlu1b expressed in Escherichia coli was electrophoretically distinct from TaGlu1a and TaGlu1c. Furthermore, coexpression of TaGlu1a and TaGlu1b gave seven bands on a native-PAGE gel, indicating the formation of both homo- and heterohexamers. One distinctive property of the wheat and rye glucosidases is that they function as hexamers but lose activity when dissociated into smaller oligomers or monomers. The crystal structure of hexameric TaGlu1b was determined at a resolution of 1.8 angstrom. The N-terminal region was located at the dimer-dimer interface and plays a crucial role in hexamer formation. Mutational analyses revealed that the aromatic side chain at position 378, which is located at the entrance to the catalytic center, plays an important role in substrate binding. Additionally, serine-464 and leucine-465 of TaGlu1a were shown to be critical in the relative specificity for DIMBOA-glucose (2-O-b-D-glucopyranosyl-4- hydroxy-7-methoxy-1,4-benzoxazin-3-one) over DIBOA-glucose (7-demethoxy-DIMBOA-glucose).

    Oat leaves produce phytoalexins, avenanthramides, in response to infection by pathogens or treatment with elicitors. The metabolism of avenanthramides was investigated using low molecular weight, partially deacetylated chitin as an elicitor. When oat leaf segments are floated on the elicitor solution, avenanthramides accumulate in the solution. The transfer of elicited oat leaves to solutions containing stable-isotope-labeled avenanthramides resulted in a rapid decrease in the labeled avenanthramides, suggesting the metabolism of avenanthramides. The rate of decrease was enhanced by elicitor treatment, and was dependent on the species of avenanthramides, with avenanthramide B decreasing most rapidly. The rates of biosynthesis and metabolism of avenanthramides A and B were measured using a model of isotope-labeling dynamics. Avenanthramide B was found to be more actively biosynthesized and metabolized than avenanthramide A. Radiolabeled avenanthramide B was incorporated into the cell wall fraction and 99% of incorporated activity was released by alkaline treatment. Gel filtration indicated that high-molecular-weight compounds derived from avenanthramide B were released by alkaline treatment. The decrease in stable-isotope-labeled avenanthramides was suppressed by catalase, salicylhydroxamic acid, and sodium ascorbate, suggesting the involvement of peroxidase in the metabolism. Consistent with this, peroxidase activity that accepts avenanthramide B as a substrate was induced in apoplastic fractions by elicitor treatment. The appearance of multiple basic isoperoxidases was observed by activity staining with 3-amino-9-ethylcarbazole coupled with isoelectric focusing of proteins from elicitor-treated leaves. These findings suggest that accumulated avenanthramides are further metabolized in apoplasts in oat leaves by inducible isoperoxidases.

    Avenanthramides, a series of substituted cinnamic acid amides with anthranilate, are phytoalexins in oats (Avena sativa L.). The precursors of avenanthramides, cinnamate and anthranilate, are biosynthesized via the shikimate pathway that branches at chorismate. Chorismate mutase (CM, EC 5.4.99.5) is the first enzyme on the branch that provides the cinnamate part of avenanthramides. The induction of CM was investigated in primary oat leaves using oligo-N-acetylchitooligosaccharides as elicitors. The CM activity started to increase 6 h after elicitation, and reached a maximum by 9 h, being around twice as large as that in control leaves. Among the oligo-Nacetylchitooligosaccharides tested, tetra-, penta-, and hexasaccharides effectively induced the CM activity in a dose-dependent manner. The activity was separated into two major peaks on anion exchange chromatography with Mono Q, indicating that at least two CM isoforms are present in oat leaves. A comparison of elution profiles of CM activity in intact and elicitor-treated leaves revealed that only one CM isoform is responsive to the elicitor. Two CM isoforms in oat leaves were partially purified and characterized. Both CM isoforms were insensitive to L-phenylalanine, L-tyrosine, L-tryptophan, and caffeate.     Oat phytoalexins, avenanthramides, are a series of substituted hydroxycinnamic acid amides with anthranilate. The anthranilate in avenanthramides is biosynthesized by anthranilate synthase (AS, EC 4.1.3.27). Induction of anthranilate synthase activity was investigated in oat leaves treated with oligo-N-acetylchitooligosaccharide elicitors. AS activity increased transiently, peaking 6 h after the elicitation. The induction of activity was dependent on the concentration and the degree of polymerization of the oligo-N-acetylchitooligosaccharide elicitor. These findings indicate that the induction is part of a concerted biochemical change required for avenanthramide production. The elicitor-inducible AS activity was strongly inhibited by L-tryptophan and its analogues including 5-methyl-DL-tryptophan, and 5- and 6-fluoro-DL-tryptophan, while the activity was not affected by D-tryptophan. The accumulation of avenanthramide A was also inhibited by treatment of elicited leaves with these AS inhibitors, indicating that a feedback-sensitive AS is responsible for the avenanthramide production. In elicited leaves, the content of free anthranilate remained at a steady, low level during avenanthramide production. Moreover, administration of anthranilate to elicited oat leaves resulted in an enhanced avenanthramide accumulation. AS may play a role as a rate-limiting enzyme in the biosynthesis of avenanthramides.

    Both N-p-coumaroyl-and N-feruloyltyramine accumulated in response to wounding in leaf segments of maize. The amount of N-hydroxycinnamoyltyramines started to increase 3-6 h after wounding and peaked at 12h. Thereafter, the amount of N-p-coumaroyltyramine decreased rapidly, while the N-feruloyltyramine content remained at a high level. The accumulation of N-hydroxycinnamoyltyramines was accompanied by an increase in the tyramine N-hydroxycinnamoyltransferase (THT) activity. This increase was initially detected 3h after wounding and reached a maximum at 36h, the level of activity being 40 and 11 times that in the leaves before wounding and in the control leaves, respectively. Partial purification of THT from wounded leaves by (NH_4)_2SO_4 precipitation and subsequent two steps of anion-exchange chromatography resulted in a 12.5-fold increase in specific activity. Kinetic studies with this partially purified enzyme revealed that the best substrates were tyramine and feruloyl-CoA, although tryptamine and sinapoyl-CoA also efficiently served as substrates. The apparent native molecular weight of the enzyme was determined by gel filtration as 40 kDa.

    Oat phytoalexins, avenanthramides, occur as constitutive components in seeds. The amounts of each avenanthramide were analyzed. The composition of avenanthramides in dry seeds was different from that in elicitor-treated leaves. In seeds, avenanthramide C was most abundant with an amount two times larger than that of avenanthramide A or B. On the other hand, avenanthramide A was the major component in elicitor-treated leaves. The total amount of avenanthramides in seeds increased 2.5 times during imbibition for 48 h although the composition did not change. The hydroxycinnamoyl-CoA:hydroxyanthranilate N-hydroxycinnamoyltransferase (HHT, EC 2.3.1.-) activity, which is responsible for the final condensation step in the avenanthramide biosynthesis, was detected in dry seeds. The activity was localized in endosperm and scutellum, and slightly increased during 48-h imbibition. The enzyme was partially purified by anion exchange chromatography from both dry seeds and elicitor-treated leaves. The activity was separated into two peaks by chromatography, indicating that HHT consists of at least two isoforms. The substrate specificities of HHT isoforms from seeds were different from each other.

    An assay method for hydroxycinnamoyl-CoA: hydroxyanthranilate N-hydroxycinnamoyl transferase (HHT) in oat leaves (Avena sativa L.), which is thought to be one of the key enzymes for the biosynthesis of avenanthramides, phytoalexins in this plant, was established. HHT activity was induced by treating the leaves with oligo-N-acetylchitooligosaccharides. Among the chitooligosaccharides tested, penta-N-acetylchitopentaose ((GlcNAc)(5)) was the most effective in inducing activity. The induction by (GlcNAc)(5) was dose-dependent, in which case HHT activity was initially detected after 6 hr and reached a maximum by 12 hr. All of the putative precursors of avenanthramides acted as substrates for HHT, with 5-hydroxyanthranilic acid and feruloyl-CoA being the best substrates for the anthranilic moiety and the cinnamoyl moiety of avenanthramides, respectively.     Victorin C, a host-specific toxin produced by Helminthosporium victoriae, induced hydroxycinnamoyl-CoA:hydroxyanthranilate N- hydroxycinnamoyltransferase (HHT, EC 2.3.1) activity in oat leaves (Avena sativa L., a cultivar carrying Pc-2 gene). This enzyme activity catalyzes the final step of biosynthesis of oat phytoalexins, avenanthramides. The HHT activity was detected after 12 h of victorin C application and reached to a maximum by 18 h. The induction of HHT was dose-dependent. All of the putative precursors of avenanthramides acted as substrates for HHT. These findings indicate that the accumulation of avenanthramides by victorin C treatment is due to induction of HHT. The enzyme activity showed highest specificity to 5-hydroxyanthranilate for the anthranyl moiety, while feruloyl-CoA was most effective for cinnamoyl moiety. HHT induced by victorin C showed significantly lower affinity for anthranilic acid relative to the enzyme induced by oligo-N-acetylchitooligosaccharides, another elicitor, suggesting that isozymes of HHT occur in this plant.

    紀の川流域の地域遺伝資源である薬用紫蘇の1系統について、外観特性および機能性成分含有量に関する遺伝を調査した結果、本系統は形態的特徴および機能性成分含有量に関して固定していると考えられた。一方、この高機能性シソの機能性向上と周年生産技術の確立を目指して、単色光補光による生育調節を試みたところ、青色光補光が花成を顕著に抑制し、ペリルアルデヒド含有量が高く維持する効果をもつことが明らかとなった。
