    Koan Ogata (1810-63), the director of Tekijuku, was a physician who contributed to the progress of Western medicine late in the Edo period. We found many kinds of crude drugs left in his medicine chest. In order to identify the origin of the drug labeled as dobuku we thoroughly investigated the medical literature published during the 16th to 20th centuries, and morphologically analyzed the contents of dobuku by making comparisons with the specimens inherited by Osaka University using a digital microscope. The literature investigation showed that dobuku means dobukuryo, and its botanical origin during the Edo period was Smilax glabra Roxb. We compared the contents of dobuku morphologically with the specimens of S. glabra and S. china L. We found that the contents have a internal morphological structure similar to that of S. glabra, which is the candidate of the botanical origin of the contents.Therefore, we believe that dobuku is dobukuryo, and its botanical origin is S. glabra. We also surveyed the clinical application of dobukuryo in medical literature and found that it was used for treating patients with syphilis during the Edo period.

    Koan Ogata (1810-63), the director of Tekijuku, was a physician who contributed much to the medical profession through his knowledge of Western medicine late in the Edo period. The medicine chest he used during the prime years of his life included many yet unknown crude drugs. In order to identify the drug labeled as saru, we conducted a thorough investigation of medical literature published during the 17th to 20th centuries. As the result of pharmacognostical analysis, we clarified the morphological characteristics of sarsaparilla, which is a candidate for the origin of the contents. We then examined specimens of Mexican and South American sarsaparillas, which were collected by E. Merck and Eli Lilly & Co. at the beginning of the 20th century, using a digital microscope. We found that these specimens showed three morphological features : 1) They had cylindrical roots with diameters of approximately 5 mm, 2) their surfaces were brown with deep vertical crinkles, and 3) they had light brown cortices and white steles covered with a creamy yellow endodermis. The contents of saru showed a high similarity to the characters of sarsaparilla written in literature and the morphological features of historical specimens. We identified them as the roots of genus Smilax, including Smilax medica and S. officinalis.

    Paeoniae Radix is one of the crude drugs frequently used in traditional Japanese medicine (Kampo medicine). It takes abundant labor and time to cultivate Paeonia lactiflora for medicinal use high production cost is one of the main reasons why the domestic production of Paeoniae Radix is decreasing in Japan. To promote the production of Paeoniae Radix, we focused on Paeonia cultivars that produce commercially valuable flowers and investigated their possibility for medicinal use. We prepared 28 batches of peony roots derived from P. lactiflora, which were cultivated in Japan 4 batches were crude drug samples, and 24 batches were cultivar roots. The elements contained in these samples were measured using inductively coupled plasma (ICP)-MS. The obtained data were then analyzed by principal component analysis (PCA) and back propagation artificial neural network (BPANN) analysis. No significant differences were found between the profiles of elements contained in crude drugs and cultivar roots. However, PCA results indicated a high similarity of the multielement fingerprints of crude drugs. Using the PCA results, we also assessed visible cluster trends and found that 5 batches of cultivars also showed fingerprints related to those of crude drugs. We certified this classification by BPANN. From the perspective of metallomics, our findings suggest that these 5 batches of Paeonia cultivars could be alternatives to crude drugs.

    The heart undergoes a major metabolic transition after birth, a change largely caused by alterations in substrate availability, hormone levels and transcription factor content. However, another factor that could contribute to the resulting upregulation of oxidative metabolism is the increase in taurine levels. We proposed that by increasing UUG decoding and the biosynthesis of mitochondria encoded proteins, elevations in taurine content enhance electron transport flux and increase oxidative metabolism. To test our hypothesis, the effect of reduced taurine content on oxidative metabolism of myocardial mitochondria and neonatal cardiomyocytes was examined. Taurine deficient neonatal mitochondria exhibited impaired oxidation of complex I specific- but not complex II specific-substrates, indicating that taurine deficiency regulates complex I activity. Taurine deficiency also reduced respiration of neonatal cardiomyocytes oxidizing carbohydrate (glucose, lactate and pyruvate). However, cardiomyocytes from 2–3 day-old hearts respiring either β-hydroxybutyrate, an important substrate in the neonatal heart, or palmitate, which is poorly metabolized during the early neonatal period, were resistant to the metabolic defects of taurine deficiency, These data support the hypothesis that taurine contributes to development of respiratory chain function after birth, which is required for oxidative metabolism of multiple substrates.

    Longgu (Fossilia Ossis Mastodi) is a non-botanical crude drug, defined as “the ossified bone of large mammal” in the Japanese Pharmacopoeia sixteenth edition (JP16). It is a non-reproducible drug and is now facing the threat of exhaustion. To solve this problem, we aimed to clarify the role of longgu in Kampo prescriptions, which has not yet been scientifically ascertained. In this study, we focused on decoction of Keishikaryukotsuboreito (KRB). The profile of inorganic and organic components in the extract was analyzed by inductively coupled plasma mass spectrometry (ICP-MS) and gas chromatography flame ionization detection (GC/FID), respectively. Twenty-five elements were detected by ICP-MS in KRB and longgu-free KRB (KB) decoctions. However, 23 elements were detected in unadultrated longgu (R) decoctions, and their total amount was 30 times lower than those of the KRB and KB decoctions were. No organic compounds were detected in R decoctions by GC/FID, though many were detected in KRB and KB decoctions. KRB decoctions were distinguished from KB decoctions by multivariate analysis. The only difference in the crude drugs was the presence of longgu, and therefore the difference in the profiles was attributed to the effect of longgu. Longgu was submitted to terahertz (THz) wave spectrometry after the decocting process. The THz spectra indicated that longgu adsorbed compounds during the KRB decoction. These results suggested that longgu not only releases its components, but also adsorbs ingredients from other crude drugs during decoction.

    Hyper-beta-alaninemia is a rare metabolic condition that results in elevated plasma and urinary β-alanine levels and is characterized by neurotoxicity, hypotonia, and respiratory distress. It has been proposed that at least some of the symptoms are caused by oxidative stress however, only limited information is available on the mechanism of reactive oxygen species generation. The present study examines the hypothesis that β-alanine reduces cellular levels of taurine, which are required for normal respiratory chain function cellular taurine depletion is known to reduce respiratory function and elevate mitochondrial superoxide generation. To test the taurine hypothesis, isolated neonatal rat cardiomyocytes and mouse embryonic fibroblasts were incubated with medium lacking or containing β-alanine. β-alanine treatment led to mitochondrial superoxide accumulation in conjunction with a decrease in oxygen consumption. The defect in β-alanine-mediated respiratory function was detected in permeabilized cells exposed to glutamate/malate but not in cells utilizing succinate, suggesting that β-alanine leads to impaired complex I activity. Taurine treatment limited mitochondrial superoxide generation, supporting a role for taurine in maintaining complex I activity. Also affected by taurine is mitochondrial morphology, as β-alanine-treated fibroblasts undergo fragmentation, a sign of unhealthy mitochondria that is reversed by taurine treatment. If left unaltered, β-alanine-treated fibroblasts also undergo mitochondrial apoptosis, as evidenced by activation of caspases 3 and 9 and the initiation of the mitochondrial permeability transition. Together, these data show that β-alanine mediates changes that reduce ATP generation and enhance oxidative stress, factors that contribute to heart failure.

    Taurine is a β-amino acid found in high concentrations in excitable tissues, including the heart. A significant reduction in myocardial taurine content leads to the development of a unique dilated, atrophic cardiomyopathy. One of the major functions of taurine in the heart is the regulation of the respiratory chain. Hence, we tested the hypothesis that taurine deficiency-mediated defects in respiratory chain function lead to impaired energy metabolism and reduced ATP generation. We found that while the rate of glycolysis was significantly enhanced in the taurine-deficient heart, glucose oxidation was diminished. The major site of reduced glucose oxidation was pyruvate dehydrogenase, an enzyme whose activity is reduced by the increase in the NADH/NAD+ ratio and by decreased availability of pyruvate for oxidation to acetyl CoA and changes in [Mg2+]i. Also diminished in the taurine-deficient heart was the oxidation of two other precursors of acetyl CoA, endogenous fatty acids and exogenous acetate. In the taurine-deficient heart, impaired citric acid cycle activity decreased both acetate oxidation and endogenous fatty acid oxidation, but reductions in the activity of the mitochondrial transporter, carnitine palmitoyl transferase, appeared to also contribute to the reduction in fatty acid oxidation. These changes diminished the rate of ATP production, causing a decline in the phosphocreatine/ATP ratio, a sign of reduced energy status. The findings support the hypothesis that the taurine-deficient heart is energy starved primarily because of impaired respiratory chain function, an increase in the NADH/NAD+ ratio and diminished long chain fatty acid uptake by the mitochondria. The results suggest that improved energy metabolism contributes to the beneficial effect of taurine therapy in patients suffering from heart failure.

    The major impetus behind the rise in energy drink popularity among adults is their ability to heighten mental alertness, improve physical performance and supply energy. However, accompanying the exponential growth in energy drink usage have been recent case reports and analyses from the National Poison Data System, raising questions regarding the safety of energy drinks. Most of the safety concerns have centered on the effect of energy drinks on cardiovascular and central nervous system function. Although the effects of caffeine excess have been widely studied, little information is available on potential interactions between the other active ingredients of energy drinks and caffeine. One of the active ingredients often mentioned as a candidate for interactions with caffeine is the beta-Amino acid, taurine. Although taurine is considered a conditionally essential nutrient for humans and is thought to play a key role in several human diseases, clinical studies evaluating the effects of taurine are limited. However, based on this review regarding possible interactions between caffeine and taurine, we conclude that taurine should neutralize several untoward effects of caffeine excess. In agreement with this conclusion, the European Union's Scientific Committee on Food published a report in March 2003 summarizing its investigation into potential interactions of the ingredients in energy drinks. At the cardiovascular level, they concluded that "if there are any interactions between caffeine and taurine, taurine might reduce the cardiovascular effects of caffeine." Although these interactions remain to be further examined in humans, the physiological functions of taurine appear to be inconsistent with the adverse cardiovascular symptoms associated with excessive consumption of caffeine-taurine containing beverages. © 2014 Springer-Verlag Wien.

    We aimed to establish a methodology for identifying Paeonia samples based on metallomic analysis. We prepared 66 batches of samples (16 batches of crude drugs and 50 batches of cultivars, comprising 64 batches of Paeonia lactiflora and 2 batches of P. veitchii) collected from Japan and China (Inner Mongolia and elsewhere) between 1996 and 2008. P. lactiflora samples were genetically classified into white peony root (WPR) type and red peony root (RPR) type. Up to 47 elements were measured by inductively coupled plasma-mass spectrometry, and RPR type crude drug samples contained up to five times as much calcium as the others. Principal component analysis (PCA) of the multi-element fingerprints obtained suggested that P. veitchii, which grows wild, were distinguishable from the other cultivated P. lactiflora samples. This was confirmed perfectly by soft independent modeling of class analogy (SIMCA). The PCA of the fingerprints of P. lactiflora crude drug samples also suggested that it was possible to classify them by production area (Japan, Inner Mongolia, and China excluding Inner Mongolia) and genetic type (RPR and WPR types). They were also classified 100 % to the predicted class by SIMCA in both cases. These analyses were successful among the samples whose collection dates varied. This simple metallomic method is an efficient approach for verifying the complex origin of Paeoniae Radix. © 2013 The Japanese Society of Pharmacognosy and Springer Japan.

    Calculus Bovis ( C . Bovis ) is a commonly used animal-derived therapeutic preparation. To meet the increasing clinical demand for the preparation, two artificial substitutes for Bos Taurus have been introduced in China: artificial C . Bovis and in vitro cultured C . Bovis . However, information on their efficacy and safety is inadequate. Therefore, we investigated the biological differences between the commonly used natural preparation and its two substitutes, with the aim of not only identifying the differences but also providing a procedure to distinguish between the different preparations. In the study, we prepared 9 natural C . Bovis, 2 artificial C . Bovis, and 2 in vitro cultured C . Bovis preparations for evaluation. Differences were noted between the three preparations relative to their effect on viability of cardiacfibroblasts from 1-dayold Wistar rats. Although natural C . Bovis had no effect on cell viability, 1-h treatment of the cells with 0.25 mg/ml of the substitutes significantly reduced cell viability, as detected by the MTS assay. Based on liquid chromatography and inductively coupled plasma mass spectrometry, the preparations also differed in composition. Indeed, the substitutes contained more taurine, cholic acid, iron, magnesium, and calcium than the natural preparations. They also differed spectroscopically. The present results reveal significant biological differences between natural C . Bovis and two of its substitutes. Since the substitutes appear to contain more taurine, cholic acid, and elements, these constituents may serve as markers to distinguish between natural C . Bovis and its substitutes. © Springer Science+Business Media New York 2013.

    We systematically organized antique crude drug specimens collected and stored at Osaka University since early 1900s in order to prepare to provide them for various crude drug studies. We utilized those specimens on the comparative morphological studies and the preliminary study of novel non-destructive analysis for the contents of Ogata Koan’s medicine chest, one of the most precious inheritances of Osaka University. In addition, we used those specimens as the morphological standards of high-quality crude drugs for the studies of domestic crude drug production and the establishment of the crude drug standards, and showed the significance of preservation and inheritance of those specimens. We also comprehensively investigated the traditional way of cultivation and processing of medical plants from the documents written after Edo period in order to provide the basic information for current domestic crude drug production projects.

    Koan Ogata was the physician who contributed to the treatment of infectious diseases at the end of the Edo period. We investigated his documents and inheritances from interdisciplinary view points and considered his medicinal strategies. We used historical crude drug samples stored in Osaka University for comparative analysis of ones left in Koan Ogata’s medicine chest. We also comprehensively investigated medical literature mainly written in Edo period and identified some of the contents of his medicine chest. What is more, we suggested that Koan applied both Western medicine which was the latest knowledge at that time and traditional Kampo medicine to provide the best treatment by statistical analysis of data collected in this project. We inspected the future preservation plan through the inspection of current insect damage and aged deterioration of Koan Ogata’s medicine chest and other medical inheritances.
