    During fingerling production of Pacific bluefin tuna (PBF) Thunnus orientalis, heavy mortality can occur immediately after juveniles are transferred from nursery tanks to sea cages; however, nighttime lighting can moderate this mortality. Additionally, various live prey aggregate due to nighttime lighting in practical sea cage culture of PBF. Here, we investigated whether the growth and viability of PBF juveniles could be improved through promoting feeding on live prey that aggregate under nighttime lighting. Two treatment groups were established using land-based tanks under constant environmental conditions, one in which the juveniles were fed live prey at night (night-feed in four replicate tanks) and the other in which juveniles were not fed during the night (control in four replicate tanks). Although the survival rate did not differ significantly between the two groups, growth was significantly improved in the night-feed group, in which 69–78% of the juveniles showed evidence of feeding during the night. Thus, nighttime lighting plays a vital role in the aggregation of various live prey. PBF juveniles consume these prey in sea cages, which promotes their growth. This may partially serve as a countermeasure against the heavy mortality observed in sea cages.

    It is important to understand the physiological stress responses in juvenile Pacific bluefin tuna (PBF), Thunnus orientalis, when transporting them from land-based tanks to sea cages, to develop countermeasures against mass mortality and enhance survival. We investigated the estimated time required to recover from transportation stress in PBF juveniles (32 and 35 days post hatch (DPH)) transported over two different distances (Exp 1; long distance (15.2 km), Exp 2; short distance (2.3 km)). Additionally, we examined the effects of simulated transport in fish that were transported from tank-to-tank, as well as starvation tolerance to determine the causes of stress responses during transportation. Whole-body cortisol levels and HIF-1 alpha expression levels in gills increased from pre-transport levels shortly after juveniles were released into the sea cage in Exp 1, but recovered within 24 h. Simultaneously, whole-body glucose levels fell over the 48 h after transport. Gene expression of the Hsp70 family in the liver was upregulated 48 h after fish were released into the sea cages in Exp 1, and was significantly upregulated after 48 h in Exp 2. These results indicated that 72 h were required for PBF juveniles to recover from transportation to sea cages. The starvation experiment suggested that PBF juveniles were significantly affected after 2 days of fasting and that the environmental conditions in the sea cage were the main factors resulting in a decrease in whole-body glucose levels and upregulation of Hsp70-family gene expression. Furthermore, changes of proximate compositions indicated that PBF juveniles consumed crude lipids and proteins, which were significantly affected by fasting for 1-2 days. We suggested that the first 3 days after stocking are a critical period for recovery from transportation stress and it is necessary to develop appropriate management techniques to alleviate the high mortality caused by these stress responses.

    Success of swim bladder inflation (SBI) is crucial for early survival of Pacific bluefin tuna (PBF) larvae, because it reduces larval sinking death by enhancing buoyancy. In Experiment 1, we examined the effect of photoperiod on SBI and survival in PBF larvae by comparing photoperiods of 9L: 15D (9L), 14L: 10D (14L: natural photoperiod), 19L: 5D (19L) and 24L: 0D (24L) during 2-10days post hatch (dph). In Experiment 2, the combined effects of photoperiod (24L and 14L) and nighttime aeration rate (enhanced night-time aeration: ENA of 1300mLmin(-1) as a countermeasure for sinking death and 130mLmin(-1)) on the survival and SBI were also examined during 2-10 dph. Moreover, in Experiment 3 the effect of photoperiod on vertical distribution of larvae in night-time was examined on 3-5 dph. Photoperiod of 24L in Experiment 1 significantly inhibited SBI compared with 14L and 19L; nevertheless, it significantly improved survival compared with other photoperiods with a dark period. On the other hand, the shortened light period (9L) showed significantly reduced SBI and also survival. In Experiment 2, the countermeasure for sinking death of ENA under 24L did not further improve the survival; rather it tended to reduce the survival. In Experiment 3, larvae distributed less in the bottom layer in 24L than in 14L, suggesting the reducing effect of 24L on sinking death. The results indicate that 24L without ENA is suitable for survival which is the most serious problem in PBF larviculture.

    Here, to evaluate ontogenic changes in visual sensitivity in juvenile longtooth grouper Epinephelus bruneus, we investigated opsin gene sequences and expression in larvae and juveniles. The longtooth grouper has 10 opsin genes: Rh1, five Rh2 genes (Rh2A1-1, Rh2A1-2, Rh2A2, g6738, and g6740), SWS1, two SWS2 genes (SWS2A1 and SWS2A2), and LWS. To date, five or six Rh2 opsin genes have been reported in perciform fishes. Differential opsin gene expression was examined by RT-PCR; the results suggested that switching from SWS1 to SWS2 occurs during early development, and that there is an absence of LWS expression between 14 and 40 days post-hatch. These ontogenetic changes in opsin gene expression may correspond to different requirements at different life stages.

    Electroretinogram (ERG) recordings and short-wave sensitive 1 (SWS1: ultraviolet sensitive) opsin gene expression data were obtained to investigate the spectral sensitivity and scotopic luminous sensitivity of juvenile longtooth groupers Epinephelus bruneus. The ERG showed broad spectral sensitivity (350-600nm; (max)=560nm approx.) in the photopic condition. The scotopic vision was maximally sensitive to green light ((max)=500nm approx.). SWS1 opsin gene expression was measured in the eyes using the reverse transcription-polymerase chain reaction method (RT-PCR). This indicated that the ultraviolet sensitivity in the photopic spectral response curve (360nm approx.) was attributable to SWS1 opsin absorption. The contribution of the -band cannot rule out. Scotopic luminous sensitivity in this species similar to that in other coastal fishes and higher than that in pelagic fishes. These results suggest that the vision of a juvenile longtooth grouper is adapted to conditions in shallow coastal waters.

    This study examined the influence of swimbladder inflation (SBI) failure on mortality (Experiment 1), lordotic deformity (Experiment 2) and growth (Experiment 1, 2) in Pacific bluefin tuna, Thunnus orientalis, postflexion larvae and juveniles by producing larvae lacking inflated swimbladders. Experiment 1 was conducted for postflexion larvae to juveniles (from 18 to 30days-post-hatch; dph). Mortality was not significantly different between the fish with (WIS) and without (WOIS) inflated swimbladders. Standard length (SL) and body weight (BW) were significantly smaller in WOIS than WIS. Moreover, the SBI was found in WOIS after postflexion stage. In Experiment 2, two examination trials were conducted on SBI, vertebral deformity and growth for juvenile stage. Lordotic deformities were found neither in the WOIS nor in the WIS. Although SL and BW were significantly smaller in WOIS than WIS at 22dph, after 37dph no significant differences were found between them. The results of Experiment 1 and 2 indicate that SBI failure causes growth retardation until juveniles of 30dph; however, it does not cause growth retardation in juveniles after 37dph and mortality of postflexion larvae and juveniles. Moreover, SBI failure did not cause lordotic deformity in PBT differently from many other fish.

    This study investigated the optimal timing of day to promote initial swimbladder inflation (ISI) for improved Pacific bluefin tuna (PBT), Thunnus orientalis, larval survival. Larval swimbladder inflation frequency was compared based on three experiments using different time schemes of surface film removal (SFR) from 3 to 9days post hatch (dph). SFR was conducted from 05:00 to 19:00hours (light period: S.5-19), 19:00 to 05:00hours (dark period: S.19-5), 08:00 to 19:00hours (S.8-19) and the entire day (S.24) in Experiment 1; from 08:00 to 19:00hours (S.8-19-E2), 08:00 to 13:00hours (S.8-13), 13:00 to 19:00hours (S.13-19) in Experiment 2; and from 13:00 to 16:00hours (S.13-16), 16:00 to 19:00hours (S.16-19), 18:00-19:00hours (S.18-19) in Experiment 3. The swimbladder inflation frequency at the experiment termination (9dph) was significantly higher (P<0.001) in S.24 (91.1 +/- 5.7%), S.5-19 (92.2 +/- 5.1%) and S.8-19 (93.3 +/- 3.4%) than in S.19-5 (11.1 +/- 5.1%) in Experiment 1, and remarkably higher in S.8-19-E2 (81.7%) and S.13-19 (88.3%) than in S.8-13 (0.0%) in Experiment 2, and significantly higher (P<0.001) in S.16-19 (84.4 +/- 5.1%) and S.18-19 (70.0 +/- 12.0%) than in S.13-16 (7.8 +/- 3.9%) in Experiment 3. These results suggest that the optimal timing to promote larval ISI by SFR is a few hours before the end of light period (16:00-19:00hours) from 3 to 9dph.

    Juvenile Pacific bluefin tuna (Thunnus orientalis; PBT) often experience high mortality during ship transportation. This study investigated whether the addition of colours or patterns to the walls of tanks affected survival rate. In the first experiment, three colours and lattice patterns were tested: dark blue single-colour, red single-colour, and red-blue lattice pattern. Fish in all tanks exhibited abnormal behaviours when sunlight entered the tanks between 0800 and 1000hours, but mortality only increased in the single-coloured tanks as a result of collision with the tank walls. In the second experiment, four colours and patterns were tested: dark blue single-colour, red-blue lattice pattern, red-blue lattice pattern with shade sheet and red-green lattice pattern with shade sheet. Again, we visually observed that fish in all treatment groups exhibited abnormal behaviour when sunlight entered the tanks, but there were no collision deaths in the lattice-patterned tanks and survival in this group was significantly higher than in the single-coloured tanks. Thus, the use of a high-contrast colour pattern can prevent mass death of juvenile PBT during ship transportation.

    To investigate the reason for the high mortality of cultured juvenile Pacific bluefin tuna (PBT), especially during land-based culture, we examined the effect of the rate of light-intensity change during the dawn period. Juvenile PBT kept in a natural light environment (control) were compared with those exposed to an artificially slowed change in light intensity during the dawn period (test group). For the test group, lamps, which were connected to automatic timers and placed above the rearing tank, were switched on from 0345 to 0515 h at 15-min intervals. After 9 d, the survival rate of the test and control group was identical. Between Day 1 and Day 6, whole-body cortisol levels were not different between groups. However, whole-body glucose levels in the test group were significantly higher than those in the control group. Both groups displayed a diel rhythm in plasma cortisol concentration that peaked at 0300-0600 h. These results suggest that slowing the increase in light intensity during the dawn period did not improve survival and that low light intensity itself induces high mortality in cultured PBT juveniles.

    We have previously reported that night-time lighting prevents the mass death of Pacific bluefin tuna (PBT) juvenile due to collision and/or contact with the walls of sea cages, immediately after transfer to the cages, and that night-time lighting does not negatively impact fish stress levels. However, the period of night-time lighting should be limited to minimize negative impacts on the surrounding environment and aid management. Therefore, we investigated the optimal period of night-time lighting by evaluating the whole-body cortisol and glucose levels as stress parameters, growth performance and survival of PBT juvenile in four cages with different periods of night-time lighting (i.e. unlit, 4-day, 8-day and 12-day lighting). The results showed that almost all fish were died 1day after transfer to the unlit cage. In comparison, the other groups (4-day, 8-day and 12-day lighting) had high survival rates (92.5-96.0%) without significant difference. However, in the 4-day-lighting group, an obvious stress response was recorded on day 5, and growth performance was significantly lower. In the 8-day-lighting group, whole-body cortisol levels were slightly elevated on day 9; however, significantly elevation was not recorded on day 12. These results indicate that the recommended lighting period of night-time lighting in sea cages is 8-12days.

    This study investigated the influence of initial swimbladder inflation (ISI) failure on survival of Pacific bluefin tuna (PBT), Thunnus orientalis, larvae to prevent mass mortality due to sinking death. In Experiment 1, swimbladder inflation frequency and survival within an ISI promoted (PS) group, for which surface film of rearing water was removed, and a group without ISI promotion (NPS) were compared in 20 and 30kL tanks. The PS group demonstrated significantly higher swimbladder inflation frequency and increased survival than the NPS group within 20kL tanks at 9days post hatching (dph). Similar tendencies were observed within 30kL tanks. In Experiment 2, larval distribution and swimbladder inflation frequency in the night-time were examined through larval sampling within the upper and middle layers of the water column and tank bottom within 1.6 and 30kL tanks at 5dph. Larvae at tank bottom had higher distributional density and significantly lower swimbladder inflation frequency than those distributed in the upper and middle layers within 1.6kL tanks. Similar tendencies were observed within 30kL tanks. Results of this study suggest the improvement of larval survival due to prevention of sinking death through ISI promotion in mass-scale PBT larviculture.

    Although the survival rate of juvenile Pacific bluefin tuna Thunnus orientalis (PBT) during 30 days after stocking in sea net cages has been as low as approximately 50%, the reason for this high mortality is not clear. The dead fish were collected and counted during 30 days after stocking in a sea net cage, and the growth performance, stomach contents, and bone injury were investigated. Nearly half (47.9%) had died within the first 4 days. The total length and body weight of the dead fish were significantly smaller than those of the live fish, and the weight of the dead fish did not increase during the first 7 days. Approximately one-third of the dead fish (21.9-42.9% per day) had ingested inorganic matter such as wood or Styrofoam waste during the first 7 days, and 25.0-45.0% of the dead fish sampled per day showed poor growth. The daily ingestion rate of inorganic matter decreased dramatically from the eighth day, when the daily mortality rate decreased. The percentage of dead fish with bone injuries due to collision or contact was low (less than 15%). These results suggest that one of the causes of high mortality during several days after stocking in a sea cage was accidental ingestion of inorganic matter by juvenile PBT.     The spectral sensitivity of the fish and the suitable light wavelength range for survival and growth performance of juvenile Pacific bluefin tuna (PBT) were investigated. The spectral sensitivity peak of PBT under photopic condition was observed between 449 and 503 nm, which corresponded to their natural habitat. The fish were reared in tanks irradiated continuously with 4 kinds of light emitting diodes (LEDs). The maximum wavelength of LEDs used for the rearing experiment were 460 nm (blue), 520 nm (green), 630 nm (red), and 450-680 nm (white). There was no notable difference in survival rate among fish in the four LED groups. However, the growth of juvenile PBT was lesser under red light compared to the green and white light wavelengths. These results suggest that PBT juveniles have low sensitivity to red light because the fish are rarely exposed to the red light wavelengths under natural ocean conditions. Thus, low sensitivity to red light negatively influenced the feeding behavior and growth of PBT juveniles.

    We examined the effects of feeding conditions and size differences on the aggressive behaviour and cannibalism in Pacific bluefin tuna (PBT) Thunnus orientalis larvae. In a 24h experiment, restricted feeding alone was found to remarkably increase the frequency of aggressive behaviour, which was further elevated by differences in fish size. In a 4-day rearing experiment, while aggressive behaviour was increased by restricted feeding alone, the frequency of cannibalism did not change significantly. Although the frequency of aggressive behaviour did not increase with difference in size factor, small fish in this group gradually tended to die over 4-day period. In the restricted feeding and size difference group, large fish were observed to bite the small fish, and almost all the small fish died on the day after the start of the experiment. These results suggest that the aggressive behaviour of PBT larvae is chiefly increased by the shortage of live food; however, deaths related to cannibalism mainly occurred in small larvae and rapidly increased with food restriction and differences in fish size.

    Juvenile Pacific bluefin tuna (PBT), Thunnus orientalis, aged 30 or more days post hatching (>= 5 cm total length) are at high risk of mortality through contact or collision with tank walls. In this study, survival rates of PBT reared under three tank wall treatments (polka dot pattern, lattice pattern, and dark-green single color (control) were investigated. White tape was attached to the tank's surface to form either a polka dot or a lattice pattern. Fish were then reared in the tanks for 9 days. Fish survival rates in the lattice patterned and polka-dot patterned tanks were higher than that in the control tank. Bone and parasphenoid injury rates in the control group were significantly higher than those in the polka dot and lattice patterned treatment groups. The plasma cortisol content of juvenile PBT reared in the control group was higher than for fish reared in patterned tanks. These results suggest that juvenile PBT mortality from contact or collision with tank walls was reduced when wall visibility was improved by creating patterns on the tank walls.     Land-based cultured juvenile Pacific bluefin tuna Thunnus orientalis (PBT) have high mortality rates due to collisions or contacts with tank walls after about 30 days of hatching. To determine the effect of night-time lighting on their survival, juvenile PBT were reared under four different night-time light intensities (0, 5, 15 and 150 lx) for 9 days, followed by a 3-day observation period. High-intensity, night-time lighting (150 lx) significantly improved the survival rate (75.8% P < 0.001) compared with the unlit control group (0 lx, 64.3%). The survival rate in the high-intensity group decreased after the end of the lighting period. Lighting did not influence whole-body cortisol levels, glucose levels, or diel changes in plasma cortisol levels. In contrast, the survival rates of fish exposed to light intensities between 5 and 15 lx were slightly lower than that of the unlit control group. These results suggest that providing night-time lighting of 150 lx or higher is an effective method for reducing the mortality of cultured PBT. © 2013 John Wiley & Sons Ltd.

    The color of the red muscle of pelagic fish is used as an indicator of freshness, and sustaining a bright red color is important for maintaining the commercial value of pelagic fish. Feeding fish with antioxidant compounds can delay metmyoglobin (metMb) formation, but the process requires long-term feeding. Live cultured Japanese horse mackerel Trachurus japonicus were used in this study. After anesthetization, 2 ml of blood was drawn from blood vessels, and a parenteral solution including an antioxidant compound was injected. Browning, metMb formation, and lipid oxidation of dark muscle during chilled storage were delayed by injecting sodium l-ascorbate as an antioxidant compound in the blood vessels of live fish. © 2013 The Japanese Society of Fisheries Science.

    Although Pacific bluefin tuna is a species that relies on vision, its photopic visual function is not well known; we therefore recorded electroretinograms to investigate photopic spectral sensitivity in juveniles of this species (49-81 days post-hatch; standard length 74-223 mm). The peak spectral sensitivity wavelength was 505 nm. We estimated that two (lambda(max) = 512-515 nm and 423-436 nm) or three (lambda(max) = 512-515 nm, 423-436 nm, and 473 nm) types of cone visual pigments contribute to photopic vision; these spectral sensitivities are adapted to surface water habitats in clear ocean and coastal water.

    Initial swimbladder inflation (ISI) of Pacific bluefin tuna (PBT), Thunnus orientalis, larvae was studied to increase the survival of cultured larvae. Experiment 1 was conducted to explore promotion and inhibition of ISI under different water surface conditions; including the use of surface skimmer to remove autogenous surface substances (SS), covering the water surface with liquid-paraffin-layer (LP) and oil film (OF), and a control (non-treatment, NT). Significantly higher inflation frequency was observed in SS (62.2%) than NT (11.9%), LP (2.7%) and OF (3.9%). This indicates that ISI in PBT larvae can be promoted by removal of surface substances on rearing water which inhibit larval air gulping. Experiment 2 aimed to elucidate proper day of larval age to start skimming for promoting ISI with four different periods of oil film removal: from 3 to 8 (SF3D), 4 to 8 (SF4D), 5 to 8 (SF5D), 6 to 8 (SF6D) days-post-hatch (dph). Significant improvement in ISI frequency was observed in SF3D (80.2%) but the frequency was very poor in SF4D, SF5D, and SF6D (17.87.5%). This implies the need of oil film removal without missing a narrow window, 1 day of 3 dph, to promote ISI in practical PBT larviculture.

    In cultured juvenile Pacific bluefin tuna (Thunnus orientalis), reducing the mass deaths caused by collision or contact with tank or net walls at night is a priority for seedling production. Pacific bluefin tuna is a visually dependant species, although its scotopic vision is poor. We recorded electroretinograms to investigate the visual function with growth in the dark-adapted eyes of juvenile Pacific bluefin tuna. Peak wavelengths of spectral sensitivity [38-62 days posthatch (dph), 77-167 mm standard length (SL)] were observed between 474 and 494 nm. Visual light sensitivity has a tendency to increase slightly with growth at 28-64 dph in individuals that measured 29-175 mm SL. However, visual temporal resolution did not significantly increase with growth at 38-62 days dph in individuals that measured 77-167 mm SL. These results suggest that the mass death continues between 28 and 64 dph because of low visual function and increasing swimming speed with growth.

    Aggressive behaviour, assessed by 24-h observations, and survival rates, determined in a 2-week rearing experiment, were examined in greater amberjack, Serbia dumerili, subjected to single and multiple factors (high density, restricted feeding and diverse size). Aggressive behaviour frequency in the group subjected to both restricted feeding and size diversity (FS) was significantly higher than that in the other groups (Tukey, n = 4, P < 0.05). In the FS group, aggressive behaviour increased with the starvation time from 19 days post hatching (dph) to 23 dph (two-way analysis of variance, Tukey, n = 4, P < 0.05), but decreased gradually thereafter to 46 dph. In the rearing experiment, the survival rates for the diverse size and FS groups were lower than those of the control or restricted feeding groups (Kaplan-Meier, log-rank test, n = 100, P < 0.01). Daily mortality in all groups was the highest at 24 dph; although dead fish were usually small, disappearance due to cannibalism was not observed during the rearing period. The results indicate that aggressive behaviour in greater amberjack is induced by hunger and size diversity in the early juvenile stages (23 dph). Mass mortality during seedling production was mainly caused by injury and the death of small fish due to aggressive behaviour/cannibalism.

    Although chub mackerel (Scomber japonicus) is widely distributed all over the world, the relevance of its visual sensitivity to its ecology is not yet fully understood. We investigated spectral sensitivity in juvenile chub mackerel in the range of ultraviolet (UV) to visible light (369-652 nm) by electroretinogram (ERG) using light-emitting diodes (LEDs). Sensitivity peaked at a wavelength of approximately 482 nm in dark-adapted fish and 525 nm in light-adapted fish. A secondary sensitivity peak in the UV range at approximately 382 nm was found in both dark- and light-adapted fish. The UV sensitivity of chub mackerel may be attributable to UV transmissibility of the optical media and to the presence of a beta-band of visible light-sensitive visual pigments, and not to an alpha-band of UV visual pigments. This UV sensitivity may be useful for feeding or communication with other fishes.

    We used electroretinogram recordings to investigate visual function in the dark-adapted eyes of the juvenile scombrid fishes Pacific bluefin tuna (Thunnus orientalis) and chub mackerel (Scomber japonicus) and the carangid fish striped jack (Pseudocaranx dentex). Despite the fast swimming speed of the Pacific bluefin tuna, analysis of flicker electroretinograms showed that visual temporal resolution in this species was inferior to that in chub mackerel. Peak wavelengths of spectral sensitivity in Pacific bluefin tuna and striped jack were 479 and 512 nm, respectively. The light sensitivity of Pacific bluefin tuna was comparable to that of chub mackerel but lower than that of striped jack. The Pacific bluefin tuna may not need high-level visual function under dim light conditions in natural habitat because it is a diurnal fish. However, this low temporal resolution and light sensitivity probably explain the mass deaths from contact or collisions with net walls in cultured Pacific bluefin tuna.

    High mortality following transfer of early juvenile Pacific bluefin tuna (PBT) aged around 30 days post hatching (about 5 cm total length) from hatchery tanks to sea net cage, particularly during the first few days, is a major problem inhibiting development of large-scale culture operations. Survival rate of the fish after stocking from land tank to a small sea cage was dropped to about 50% or less by the next morning, and became about 10% thereafter. In addition, the whole body cortisol levels increased significantly the next day. In a lab-based study using an infra-red camera, death was associated with fish touching and/or colliding with the net wall, then exhibiting abnormal swimming behavior, then becoming moribund during the night and dawn. In trials in sea net cages, overhead lights providing 200-3000 Ix intensity at the water surface, significantly improved survival to 73% up to 23 days post-transfer compared to 12% survival in un-lit control sea cage. Skin injuries were observed in most of the dead fish. The scotopic visual threshold of juvenile PBT was 11.1 log photons/cm(2)/s, at least 40 fold inferior to the threshold of 8.6-9.6 log photons/cm(2)/s exhibited by four marine teleosts, juvenile Pagrus major, Takifugu rubripes, Seriola dumerili, and Epinephelus septem-fasciatus. It was thought that the PBT had scotopic vision that was much less developed than that of other marine fish in the early juvenile stage, and they touched and/or came into collision with the net wall during the night and dawn.     In bluefin tuna aquaculture, collision of juveniles with the tank or net walls is a major cause of high mortality. This problem may be related to color sensibility of the visual mechanisms of this species. As a first step in understanding of color vision of Pacific bluefin tuna Thunnus orientalis, we applied a molecular technique and histology to study cone cell distribution in the retina of juvenile fish. We isolated three cone opsin genes encoding one blue-sensitive (SWS2) and two green-sensitive (RH2) visual pigments. In situ hybridization revealed that SWS2 mRNA is localized in the single-cone photoreceptors. The localization of the two RH2 signals in distinct cone cells was not determined, probably because of the high homology between the two RH2 genes. Single-cone photoreceptors appeared frequently in the ventral-temporal retina in approximately 80-mm fish and in the temporal retina in approximately 230-mm fish. These cone distributions may define a visual field with best color contrast vision in front and above the fish with a short wavelength (blue) reflecting target (sensed by single cones), and may be enhanced against the longer wavelength (green) background when fish see a target below them (sensed by double cones).

    The hypoxic condition, which sometimes occurs in the high-density layer of fish eggs during the procedures of egg collection and transportation in aquaculture, is thought to be accompanied by hypercapnia. This study investigated the effects of hypoxia and hypercapnia for the striped jack Pseudocaranx dentex embryos during somitogenesis. The somitic disturbances in newly hatched larvae were induced by the extreme hypoxia of exposure to 0 % dissolved oxygen (DO) concentration for 30 and 60 minutes. Extreme hypercapnia of 120 mg/L dissolved carbon dioxide (DCD) concentration for 90 or 120 minutes also induced the disturbances. The moderate hypoxic condition of exposure to 25% DO concentration for 60 and 90 minutes accompanied with hypercapnia of exposure to 120 mg/L DCD concentration induced somitic disturbances whereas this hypoxic condition alone did not induce them. The incidence rate of somitic disturbances was higher in the case of hypoxia of 25% DO and hypercapnia of 120 mg/L DCD than that in the case of normoxia of 100% DO and hypercapnia of 120 mg/L. These results indicate that hypoxia and hypercapnia during somitogenesis induce somitic disturbances as in other marine fishes and their coincidence promotes the induction in striped jack.

    Changes in tolerances to hypoxia and sodium azide, an indicator of cellular respiration, and activities of various energy metabolism-related chemical components were studied in Japanese flounder Paralichthys olivaceus during its early life stages from 3.5 to 20.5 mm in total length (TL). They showed flexion stage around 10.4 min TL. Lethal levels of hypoxia increased with growth from 3.5 to 8 min total TL, and the levels remained high in larvae, until 10.4 turn TL, decreased significantly thereafter. The 50% lethal concentration of sodium azide temporarily increased at 4.5 mm TL, diminished drastically between 4.5 and 10.4 nun TL, and then increased again in post-flexion larvae. Cytochrome c oxidase activity was highest in larvae around flexion, at 10.4 min TL, and subsequently decreased. In post-flexion larvae at 13.0 min TL, lactate dehydrogenase (LDH) and creatine kinase activities increased; LDH activity decreased at the juvenile stage. The adenosine triphosphate content and energy charge in fish were consistently higher in the larval stage than in the juvenile stage. These results indicated that, from just before flexion to the post-flexion stage, the energy metabolism of larvae is higher due to activated aerobic and subsequent anaerobic metabolism for metamorphosis; as a consequence, hypoxia tolerance in fish is the lowest during the increase of aerobic metabolism just before and around flexion.     Schooling behaviour and histological retinal light adaptation in juvenile Pacific bluefin tuna Thunnus orientalis were examined under various light intensities to determine the effect of light intensity on behaviour. After monitoring the schooling behaviour of juveniles 35-36 and 45-46 days post hatching, schooling variables such as nearest neighbour distance and separation swimming index were measured under different light intensities. Furthermore, retinal indices of light adaptation were investigated histologically for each experimental light intensity. Under intensities >5 lx, schooling variables in the two juvenile growth stages were nearly constant, allowing schooling. In contrast, the schooling variables indicated that the fish gradually swam more widely and randomly with decreasing light intensities <5 lx. The retinal indices also showed a shift from light adaptation to dark adaptation at light levels <5 lx. From 5 to 0-01 lx, retinal adaptation and fish schooling behaviour changed with light intensity. These data suggest that the schooling behaviour of juvenile Pacific bluefin tuna is greatly affected by retinal adaptation.     Changes in tolerance to hypoxic stress and oxygen consumption were studied in the red sea bream, Pagrus major, from its early life stage until 42 days post-hatch. In the experiments, metamorphosis was observed mainly from days 15 to 30, and the morphological shift from larva to juvenile was completed at around 9.5 mm total length (TL). During the larval stage, lethally low dissolved oxygen (DO) levels and mass-specific metabolic rates increased with growth from 2.6 to 5 mm TL (P<0.01). Subsequently, the levels remained high and decreased until about 9.5 mm TL around the flexion stage and post flexion stage. Finally, beginning in the juvenile stage, lethal DO levels and mass-specific metabolic rates decreased as TL increased up to about 30 turn (P<0.01). The relationship between lethal DO levels and mass-specific metabolic rates was significantly linear (r=0.59,p<0.001, n=207) in fish larvae and juveniles. These results indicated that, around the stage of flexion and post flexion larvae in red sea bream, metabolic rates were highest during metamorphosis, and consequently hypoxia tolerance was lowest. It was presumed that the increasing metabolic rate during metamorphosis induced a decrease in the metabolic scope of activity and thereby induced the decrease of the tolerance to some environmental stressors in the background.     Stress-induced apoptosis was characterized in larval embryos of Japanese flounder using a caspase-3-like activity assay and terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) staining of whole mounts. Severe stress conditions, such as heat shock by temperature shift from 16degreesC to 28degreesC or 29degreesC for 1 h, and ultraviolet (UV) irradiation at 20-100 MJ/cm(2), induced extensive apoptosis in embryos. Following the induction of stress, many TUNEL-positive apoptotic cells were found in the yolk sac, heart, and larval fin. Caspase-3-like activity against a fluorogenic substrate, acetyl-Asp-Glu-Val-Asp-alpha-4-methylcoumaryl-7-amide, increased in a dose-dependent manner during stress-induced apoptosis. In addition, C2-ceramide treatment (5-10 muM) induced a similar apoptosis pattern to stress treatment. These findings indicate that a pro-apoptotic pathway, which is caspase-3 mediated and acts through ceramide signaling, plays a major role in induction of apoptosis under stress conditions.

    Changes in tolerances to certain environmental stressors, e.g., low and high temperatures and salinity as well as high concentrations of ammonia, were studied in the red sea bream Pagrus major during its early life stage up to 42 days post-hatch. Relative tolerance was assessed by determining 24-h median lethal level. In laboratory experiments, metamorphosis was observed mainly from days 14-21, and the morphological shift from larva to juvenile was indicated at 9.5 mm total length at 28 days post-hatch. Among the stages tested, larva immediately after hatching showed maximum tolerance to all stressors. Tolerances to both low and high temperatures decreased up to 14 days post-hatch and increased thereafter. Hypo- and hyper-salinity tolerances also decreased up to the 14(th) day post-hatch and then showed a gentle increase. In regard to ammonia tolerance, the 50% lethal concentration diminished drastically between days 2 and 7, showed the lowest level on day 21, and subsequently rose. These results indicate that, in red sea bream under these experimental conditions, all tolerances to temperature, salinity, and ammonia stress temporarily fell during metamorphosis from days 14-21. It is considered that the depression of the scope for activity based on increasing oxygen consumption induced the decrease in various stress tolerances during metamorphosis. It is also suggested that the 50% lethal levels of various environmental stressors can be used as a stress test to evaluate the hardiness of a hatchery-produced red sea bream.

    Nile tilapia were exposed to severe hypoxic conditions. The respiratory frequency (RF) of the fish reached a maximum level at approximately 10 mmHg PO2 and decreased gradually thereafter. The fish sank to the bottom and subsequently their respiration began to stop. The hypoxic conditions increased the hematocrit value. As RF decreased, plasma cortisol and glucose levels climbed drastically. Just before respiratory arrest, the ATP and the total adenylate (TA) concentrations were significantly depressed in the liver, kidney and ordinary muscle but not in the heart or gill. As RF decreased, ATIP and TA in both the liver and kidney decreased rapidly. Cytochrome oxidase activity increased significantly in the brain, heart, gill and ordinary muscle until respiratory frequency peaked. However, from the peak RF, this activity decreased in the liver and kidney. Lactic acid levels in both ordinary muscle and liver increased markedly when the fish sank. These results indicate that the stress response to hypoxic conditions is induced markedly with the decreased RF It is also suggested that the decreased energy status in the liver and kidney of tilapia occurs at the same stage as the metabolic depression in the whole body, without increases in the aerobic and anaerobic metabolisms.

    平均体重5.4gのイシダイ稚魚を、ピリドキシン(PN)含量が1kg乾飼料当たり0.3から50.2mgの7飼料区の精製試験飼料でそれぞれ14週間飼育した。PN0.3mg区には飼育2週目から成長低下、神経過敏、遊泳異常、へい死魚の増加、正色素性小赤血球性貧血などの欠乏症状が確認された。PN1.4mg区でも飼育11週目から食欲が減退し、増重率が低下した。両区の半数の魚にPN 11.1mg/kg飼料を与えた結果、明らかな回復が認められた。試験終了時におけるPN 0.3および1.4mg区の血漿総タンパク質、グルコース、トリグリセリド含量および比肝重は他区に比べて低かった。PN 2.9mg以上の飼料区における魚の飼育成績、血液性状、血漿化学成分および比肝重に、顕著な差異はみられなかった。さらに魚体重100g当たりの肝臓GOTおよびGPT活性はPN 2.9mg以上で、肝臓PN含量はPN 11.1mg以上の飼料区でおよそ一定となった。Broken-line regression analysisにより、成長、肝臓GOTおよびGPT活性を指標としたイシダイ稚魚のPN要求量は、飼料乾重1kg当たり2.8~4.1mg、肝臓PN飽和量を指標とした場合には、11.4mgと判定された。

    Young red sea bream, Pagrus major, were exposed to a gradual decrease in partial pressure of oxygen (Po2) down to 20 mmHg for 5 h, and then sustained under 20 mmHg Po2 for an additional 3 h at 19.9°C. Fish respiratory frequency slowly increased and attained a peak level at approximately 35 mmHg Po2, and then gradually decreased. Fish sank to the tank bottom about an hour after Po2 fell below 20 mmHg, and respiration began to arrest after another hour. Hematocrit value increased and mean cellular hemoglobin content (MCHC) decreased with increasing hypoxia load. Plasma cortisol and glucose levels were significantly augmented when respiratory frequency decreased. ATP, total adenylate (TA) concentration and energy charge (EC) in the hepatopancreas, in addition to TA in the kidney, were considerably diminished by hypoxia. However, this was not observed in the gill and brain of all fishes prior to respiratory arrest. In the hepatopancreas, levels of ATP, TA and EC notably declined as respiratory frequency decreased. These results show that the cortisol stress response to hypoxia is significantly induced with the decreased respiratory frequency. Decreased energy status in the hepatopancreas appears to be important and occurs with stress response as a metabolic depression in the whole body.

    マダイ稚魚に、酸素分圧(Po2)を151から26mmHgまで2時間30分かけて低下し、26mmHg以下で約2時間放置するストレスを27.8℃で負荷した。呼吸数は徐々に増加し、38mmHg Po2前後で最大値を示してその後に低下した。26mmHg Po2以下では横転後に呼吸停止する魚が増えた。Ht値および血漿グルコース濃度はストレスの負荷に伴って増大し、MCHCは低下した。普通筋のCK、LDH活性および乳酸含量、並びに肝膵臓の乳酸含量は横転時に増加した。鰓のシトクロムcオキシターゼ(CCO)活性に顕著な変化はなかったが、脳のそれは低酸素負荷に伴って有意に増大した。一方、肝膵臓、腎臓および普通筋のCCO活性は、呼吸数最大時に激減したが、横転時以降には回復し、心臓のCCO活性も同時期に増加した。これよりマダイは、呼吸数最大時以降に低酸素負荷に対する代謝抑制を示すが、横転後には嫌気代謝の亢進とともに、その機能は解除されることが示唆された。

    In order to realize the biological management of the devil stinger Inimicus japonicus in culture, the diurnal changes of this species' oxygen consumption was recorded using a respiration monitor with a dissolved oxygen meter, YSI-57. Twenty-four fish were used. Their initial average body weight of fish was 76.3 ± 12.0 g. The fish were starved for 2 days before examination. They were then set one by one in a respiration chamber of 750 ml for 2 to 3 days. Water was pumped into the chamber by a peristaltic pump at a rate of 110 ml/min. The inlet water and outlet water of the chamber were pumped alternately to an oxygen electrode cell every 15 minutes, which process was controlled by a timer. The dissolved oxygen contents between inlet water and outlet water for the chamber were continuously recorded on a pen-recorder. The difference was determined manually from the recordings.
    The fish showed a bi-modal diurnal changes. The first peak of oxygen consumption, 178.8 ml/kg/h, was observed at twilight at 06:00. The second peak, 174.9 ml/kg/h, was recorded 2 to 3 hours after dusk, at 21:00. Lower rates of oxygen consumption, 154.8 ml/kg/h and 149.6 ml/kg/h, were found at 03:00 and 16:00, respectively. These results suggested that the oxygen generator should be operated at least twice a day, at around twilight and dust. The average rates of oxygen consumption in resting stage and active stage were 140.1 ± 64.8 ml/kg/h and 195.1 ± 89.1 ml/kg/h, respectively. The highly relationships between oxygen consumption in active and rest in fish were obtained in both twilight (r = 0.9618 with p < 0.001) and in dusk (r = 0.9625 with p < 0.001), respectively.

    The qualitative requirements and deficiency signs of water-soluble vitamins were studied in tiger puffers Takifugu rubripes having mean body weight of 2.9 g. The qualitative requirements of tiger puffer for water-soluble vitamins indicate that the requirement of this species for vitamin B6 is lower than that of other fish species such as red sea bream, yellowtail, and Japanese parrot fish, while that for vitamins related to lipid metabolism, such as choline, nicotinic acid, pantothenic acid, biotin and inositol, is comparatively high.

    The growth, maturation and chemical composition of normal diploid and triploid Japanese parrot fish, Oplegnathus fasciatus (TEMMINCK et SCHLEGEL) reared for 3 years 5 months were compared.
    Only slight differences between diploid and triploid fish in growth were observed throughout the experimental period.
    The gonado-somatic index of female triploid fish was consistently lower than that of female diploid fish. On the other hand, that of male triploid fish was higher than that of female triploid fish, and male triploid could be spawned by hand-stripping.
    In fish in the spawning period (3 and 4 years old), the % total edible portion (as percent of total body weight) in triploid fish was significantly higher than that of diploid fish.
    The lipid content of the viscera, liver and muscle in the diploid fish decreased between the pre-spawning and spawning periods, while in triploid fish, that of the viscera and abdominal muscle markedly increased during the same period. These results suggest that the accumulation of energy associated with sterility in triploid fish manifests as increase in the lipid content of the viscera and its limbus.
    In the spawning period, the crude protein and lipid contents of dorsal muscle in 4-year-old triploid fish were higher than those in diploid fish. On meat quality test, the taste and texture of female triploid fish were assessed as better than those of the female diploid.

    Control and stressed groups of juvenile Japanese parrot fish Oplegnathus fasciatus were fed diets (moist pellet) supplemented with 0, 75, and 300 mg of ascorbic acid (AsA) per 100 g. Fish in the stressed group were intermittently exposed to decreasing water oxygen levels every 3 or 4 days for 16 weeks. In the fish given a diet containing no AsA supplement, AsA-deficiency symptoms were manifested earlier, and to a greater extent, in the stressed fish than in the non-stressed ones. Similarly, the growth of stressed fish fed on a diet supplemented with 75 mg AsA per 100 g was inferior to the growth of the control group, and in the stressed fish there was a reduction in AsA levels in the plasma, kidneys, and gills. On the other hand, fish fed on a diet supplemented with 300 mg of AsA per 100 g were barely affected by the stressor. These results indicate that, in Japanese parrot fish under these experimental conditions, exposure to intermittent hypoxic stress not only induced AsA-deficiency disease early, but also increased the AsA requirement. It was also shown that high doses of AsA increased the ability of these fish to resist the stressor.

    Juvenile Japanese parrot fish Oplegnathus fasciatus were fed purified test diets supplemented with 0, 25, 50, 75, 100 and 300 mg of ascorbic acid (AsA) per 100 g of diet. In the no AsA supplemented diet group, growth declined and deficiency signs of deformity or abnormal swimming appeared from week 8. At week 16, the values of blood characteristics such as hemoglobin, hematocrit, glucose, total protein, alkaline phosphatase and calcium, and the hepatosomatic index of the fish fed diet containing no AsA supplement had significantly decreased than the other groups. On the other hand, normal growth and regular values of hematological parameters with no deformities were observed among the fish groups fed with diets with supplementary AsA levels of 25 mg per 100 g diet or greater. Moreover, the AsA contents of the brain, gill, kidney and liver of the fish were reached saturation level when the supplementary AsA level exceeded 50 mg per 100 g of diet. These results indicate that the sufficient supplementary AsA level to maintain normal growth and health of the Japanese parrot fish was estimated to be 25 mg of AsA per 100 g of diet under this experimental condition. It was also estimated that 50 mg of supplementary AsA per 100 g of diet is needed to saturate the tissue AsA levels.

    Some indicators of environmental stressors were examined in the development of healthy fish seedling production. In addition, ontogenic changes in the stress response to environmental stressors were studied in some kinds of fish in their lifespan from larvae through the juvenile stage. At first, Pagrus major juveniles were subjected to low oxygen, high and low temperatures, hypo-and hyper-salinity. The specific activities of heat shock factor (HSF), glucocorticoid receptor (GR) and cortisol level as stress indicators in the fish were compared. The cortisol concentration was increased just after stress exposure, but the specific activities of HSF and GR gradually tended to become elevate. Fish showing both high and low tolerance to stress exposure were also selected and determined to be stress indicators. HSF and GR activities in high tolerance fish tended to be higher than those in the low tolerance fish. The HSF, GR and cortisol level were suggested to be good indicators to check in fish. Next, the ontogeny of hypoxia tolerance and stress response were studied in Pagrus major, Paralichthys olivaceus, Takifugu rubripes and Thunnus orientalis larvae and juveniles. The results indicated that around the stages of flexion and post-flexion larvae in fish, metabolic rates and/or energy metabolism were the highest, and consequently hypoxia tolerance was the lowest. Finally, ontogenic changes in some stress tolerances and the response to this were examined in larvae and juveniles. It was found that at the early juvenile stage all of the tolerances and the cortisol level were increased. The effects of cortisol immersion on various stress tolerances in larvae were also tested. The response of the survival rate, cortisol concentration and GR activity to stressors showed a tendency to increase as a result of cortisol immersion. It was therefore suggested that cortisol has an important role in adaptation to environmental stressors.

    Polyculture of fish and seaweeds is a benefical system in aquaculture. In spite of its importance from the ecological point of view, little attention has been given in polyculture system. This may be due to the fact that fish and seaweeds have different growth seasons ; fish grow in summer nd autum while seaweeds propagate in winter and spring. Previously, we found the culture techniques of sterile Ulva pertusa which grow in summer and autum as same seasons as fish growth. In the presesnt study, we have examined the polyculture of fish and seaweed, sterile Ulva pertusa. The first experiment was conducted in the fish cage culture farm from August 1993 to February 1994 at the Azumacho Fish Seedling Center. Two cages, 7x7x7m, were employed, one for polyculture experiment and the other one for control experiment by the routine method. Three years old red sea bream Pagrus major, about 1750 individuals each, were cultured in each cage. The Chlorophyta, steriled Ulva pertusa was cultured by the floating net system in the experimental cage. Dissolved oxygen and carbon dioxide were measured by Radiometer ABL-330. Considering the value of pO2 in the control cage as 100%, pO2 in the experiment cge stands 109.1%. On the other hand, average pCO2 in the control cage as 100%, pCO2 in the experiment cage stands 96.1%. From the results mentioned above, it may concluded that the pCO2 in the experimental cage decreased to 4% while pO2 augmented up to 9% compared to those in the control cage. The second experiment was also conducted in order to know the efficiences of light intensity and water temperture on growth of sterile Ulva pertusa as a foundamental study of "harmonized fish culture with environment" where substances were recycled through the alga. Tendency of low light and high light intensities were observed around the winter solstice and summer solstice, respectively. The highest value was 19.7 mol/m2/day around the winter silstice and 47.3 mol/m2/day around the summer solstice. In multiple regression analysis, multiple regression coefficient of 0.74 was obtained.
